Abstract

The invention relates to diacyl indazole derivatives of general formulae (I) or (II) and to the pharmaceutically acceptable salts thereof: Wherein X, R1 and R2 are as defined herein. The invention also relates to the use of these compounds to treat diabetes mellitus, dyslipidemia and atherosclerotic disorders.

Claims (63)

1. A compound of the formulae I or II:

wherein

X is identically or differently ═C(—R)—;

R is identically or differently hydrogen, halogen, (C 1 -C 6 )-alkyl, (C 1 -C 3 )-alkyloxy-(C 1 -C 3 )-alkylene, (C 1 -C 3 )-haloalkyl, hydroxy, (C 1 -C 6 )-alkylmercapto, amino, (C 1 -C 6 )-alkylamino, di-(C 2 -C 12 )-alkylamino, mono-(C 1 -C 6 )-alkylaminocarbonyl, di-(C 2 -C 8 )-alkylaminocarbonyl, COOR3, cyano, (C 1 -C 6 )-alkylsulfonyl, (C 1 -C 6 )-alkylsulfinyl, aminosulfonyl, nitro, pentafluorosulfanyl, phenyl, heterocycle, phenoxy, O-heterocycle, CO—NR6R7, O—CO—NR6R7, O—CO—(C 1 -C 6 )-alkylene-CO—O—(C 1 -C 6 )-alkyl, O—CO—(C 1 -C 6 )-alkylene-CO—OH, O—CO—(C 1 -C 6 )-alkylene-CO—NR6R7 or unsubstituted or mono- or poly-F-substituted (C 1 -C 6 )-alkyloxy;

R1 is identically or differently (C 5 -C 16 )-alkyl, (C 3 -C 12 )-cycloalkyl, Y-aryl, Y-heterocycle or bicycle, where cycloalkyl, aryl, heterocycle or bicycle are optionally substituted one or more times by halogen, (C 1 -C 6 )-alkyl, (C 1 -C 3 )-alkyloxy, hydroxy, (C 1 -C 6 )-alkylmercapto, amino, (C 1 -C 6 )-alkylamino, di-(C 2 -C 12 )-alkylamino, mono-(C 1 -C 6 )-alkylaminocarbonyl, di-(C 2 -C 8 )-alkylaminocarbonyl, (C 1 -C 6 )-alkyloxycarbonyl, (C 1 -C 6 )-alkylcarbonyl, cyano, trifluoromethyl, trifluoromethyloxy, (C 1 -C 6 )-alkylsulfonyl, aminosulfonyl, and nitro;

Y is (C 1 -C 3 )-alkyl, which is optionally substituted one or more times by halogen, (C 1 -C 3 )-alkyl, hydroxy or trifluoromethyl;

R2 is hydrogen; or

R1 and R2 together with the nitrogen atom carrying them form a monocyclic, saturated or partly unsaturated 4- to 7-membered ring system or a bicyclic saturated or partly unsaturated 8- to 14-membered ring system whose individual members of the ring systems are optionally replaced by one to three atoms or atomic groups from the series —CHR4-, —CR4R5-, —(C═R4)-, —NR4-, —C(═O)—, —O—, —S—, —SO—, —SO 2 —, with the proviso that two units from the series —O—, —S—, —SO—, —SO 2 — are not adjacent to each other;

R3 is hydrogen, (C 1 -C 6 )-alkyl or benzyl;

R4, R5 are identically or differently (C 1 -C 6 )-alkyl, halogen, trifluoromethyl, phenyl, heterocycle, COOR3 or (C 3 -C 12 )-cycloalkyl; and

R6, R7 are identically or differently hydrogen, (C 1 -C 6 )-alkyl, phenyl, benzyl or (C 3 -C 12 )-cycloalkyl; or

a tautomer thereof or a physiologically tolerated salt thereof,

with the proviso that not more than one R is phenyl or heterocycle.

2. The compound of the formulae I or II as claimed in claim 1 , wherein:

X is identically or differently ═C(—R)—;

R is identically or differently hydrogen, halogen, (C 1 -C 6 )-alkyl, hydroxy, (C 1 -C 3 )-alkyloxy, amino, COOR3, trifluoromethyl, (C 1 -C 6 )-alkylsulfonyl, nitro, pentafluorosulfanyl, phenyl, (C 5 -C 7 )-heterocycle, CO—NR6R7, O—CO—NR6R7 or O—CO—(C 1 -C 6 )-alkylene-CO—O—(C 1 -C 6 )-alkyl;

R1 is identically or differently (C 6 -C 10 )-alkyl, (C 6 -C 12 )-cycloalkyl, Y-phenyl or Y-heterocycle or bicycle, where cycloalkyl, phenyl, heterocycle or bicycle are optionally substituted one or more times by halogen, (C 1 -C 6 )-alkyl, (C 1 -C 3 )-alkyloxy, hydroxy, (C 1 -C 6 )-alkylmercapto, amino, (C 1 -C 6 )-alkylamino, di-(C 2 -C 12 )-alkylamino, mono-(C 1 -C 6 )-alkylaminocarbonyl, di-(C 2 -C 8 )-alkylaminocarbonyl, (C 1 -C 6 )-alkyloxycarbonyl, (C 1 -C 6 )-alkylcarbonyl, cyano, trifluoromethyl, trifluoromethyloxy, (C 1 -C 6 )-alkylsulfonyl, aminosulfonyl or nitro;

Y is —CH 2 —, which is optionally substituted once by fluorine, methyl or hydroxy;

R2 is hydrogen; or

R1 and R2 together with the nitrogen atom carrying them form a monocyclic, saturated 5- to 6-membered ring system or a bicyclic saturated or partly unsaturated 9- to 10-membered ring system, whose individual members of the ring systems are optionally replaced by one to three atoms or atomic groups from the series —CHR4-, —CR4R5-, —(C═R4)-, —NR4-, —O— or —S—, with the proviso that two units from the series —O— or —S— are not adjacent to each other;

R3 is hydrogen, (C 1 -C 6 )-alkyl or benzyl;

R4, R5 are identically or differently (C 1 -C 6 )-alkyl, halogen, trifluoromethyl, COOR3, (C 3 -C 7 )-cycloalkyl, phenyl or heterocycle; and

R6, R7 are identically or differently hydrogen, (C 1 -C 6 )-alkyl, phenyl, benzyl or (C 3 -C 12 )-cycloalkyl; or

a tautomer thereof or a physiologically tolerated salt thereof,

with the proviso that not more than one R is phenyl or (C 5 -C 7 )-heterocycle.

3. The compound of the formulae I or II as claimed in claim 1 , wherein:

X is identically or differently ═C(—R)—;

R is identically or differently hydrogen, halogen, hydroxy, (C 1 -C 6 )-alkyl, trifluoromethyl, (C 1 -C 3 )-alkyloxy or amino;

R1 is identically or differently (C 6 -C 10 )-alkyl, (C 6 -C 12 )-cycloalkyl, Y-phenyl or Y-heterocycle, where cycloalkyl, phenyl, or heterocycle are optionally substituted one or more times by halogen, (C 1 -C 6 )-alkyl, (C 1 -C 3 )-alkyloxy, hydroxy, amino, trifluoromethyl or trifluoromethyloxy;

Y is —CH 2 —;

R2 is hydrogen; or

R1 and R2 together with the nitrogen atom carrying them form a monocyclic, saturated 5- to 6-membered ring system or a bicyclic saturated or partly unsaturated 9- to 10-membered ring system, whose individual members of the ring systems are optionally replaced by one to three atoms or atomic groups from the series —CHR4-, —CR4R5-, —(C═R4)-, —NR4-, —O— or —S—, with the proviso that two units from the series —O— or —S— are not adjacent to each other;

R3 is hydrogen, (C 1 -C 6 )-alkyl or benzyl; and

R4, R5 are identically or differently (C 1 -C 6 )-alkyl, halogen, trifluoromethyl, COOR3, (C 3 -C 7 )-cycloalkyl, phenyl or (C 5 -C 7 )-heterocycle; or

a tautomer thereof or a physiologically tolerated salt thereof.

4. The compound of the formulae I or II as claimed in claim 1 wherein:

X is identically or differently ═C(—R)—;

R is identically or differently hydrogen, halogen, hydroxy, (C 1 -C 6 )-alkyl, trifluoromethyl, (C 1 -C 3 )-alkyloxy or amino;

R1 is identically or differently (C 6 -C 10 )-alkyl or Y-phenyl, which is optionally substituted by halogen, (C 1 -C 6 )-alkyl, (C 1 -C 3 )-alkyloxy or trifluoromethyl;

Y is CH 2 ;

R2 is hydrogen; or

R1 and R2 together with the nitrogen atom carrying them form a monocyclic, saturated 5- to 6-membered ring system, whose individual members are optionally replaced by one to three atoms or atomic groups from the series —CHR4-, —NR4-, —O— or —S—, with the proviso that two units from the series —O— or —S— are not adjacent to each other; and

R4 is (C 1 -C 6 )-alkyl, cyclopropyl, trifluoromethyl or phenyl; or

a tautomer thereof or a physiologically tolerated salt thereof.

5. The compound of the formulae I or II as claimed in claim 1 , wherein:

X in position 4, 5 and 7 is ═C(—R)— wherein R=hydrogen, and in position 6 is ═C(—R)—wherein R is not hydrogen.

6. A pharmaceutical composition comprising one or more compounds of the formulae I or II as claimed in claim 1 or a pharmaceutically acceptable salt thereof in combination with at least one pharmaceutically acceptable excipient.

7. A pharmaceutical composition comprising one or more compounds of the formulae I or II as claimed in claim 2 or a pharmaceutically acceptable salt thereof in combination with at least one pharmaceutically acceptable excipient.

8. A pharmaceutical composition comprising one or more compounds of the formulae I or II as claimed in claim 3 or a pharmaceutically acceptable salt thereof in combination with at least one pharmaceutically acceptable excipient.

9. A pharmaceutical composition comprising one or more compounds of the formulae I or II as claimed in claim 4 or a pharmaceutically acceptable salt thereof in combination with at least one pharmaceutically acceptable excipient.

10. A pharmaceutical composition comprising one or more compounds of the formulae I or II as claimed in claim 5 or a pharmaceutically acceptable salt thereof in combination with at least one pharmaceutically acceptable excipient.

11. A method for the treatment of diabetes mellitus comprising administering to a patient in need of said treatment a therapeutically effective amount of a compound of the formulae I or II as claimed in claim 1 or a pharmaceutically acceptable salt thereof.

12. A method for the treatment of dyslipidemia comprising administering to a patient in need of said treatment a therapeutically effective amount of a compound of the formulae I or II as claimed in claim 1 or a pharmaceutically acceptable salt thereof.

13. A method for the treatment of atherosclerotic disorders comprising administering to a patient in need of said treatment a therapeutically effective amount of a compound of the formulae I or II as claimed in claim 1 or a pharmaceutically acceptable salt thereof.

14. A process for producing a medicament comprising one or more of the compounds of the formulae I or II as claimed in claim 1 , which comprises mixing the latter with a pharmaceutically suitable carrier and converting this mixture into a form suitable for administration.

15. A process for preparing compounds of the formulae I or II as claimed in claim 1 , which comprises:

a) acylating an indazole derivative of the formula III with a carbamoyl chloride of the formula IV;

or

b) reacting in two stages initially with phosgene or an equivalent chosen from trichloromethyl chlorocarbonate, ditrichloromethyl carbonate or 4-nitrophenyl chloroformate and in a second step with an amine of formula V,

in which X, R1, R2 and R3 are as defined in claim 1 ,

and subsequently separating the compounds I and II,

16. A process for preparing compounds of the formulae I or II as claimed in claim 1 , wherein R2 is hydrogen, which comprises:

reacting an indazole derivative of the formula III with isocyanate of the formula VI: O═C═N—R1, and subsequently separating the compounds I and II.
