hT2R54 receptor polypeptides and nucleic acid sequences
Functional assays that detect the effect of a particular compound or compounds on the activation of at least one human T2R polypeptide are provided. These assays include e.g., assays which detect the effect of said compound on intracellular calcium, second messengers such as cAMP, cGMP, current e.g., by use of voltage-clamp or patch-clamp, techniques, fluorescence polarization or FRET assays, and the like. These assays are useful in identifying compounds that putatively elicit or modulate (inhibit or enhance) bitter taste in human subjects, e.g. bitter taste blockers. The effect of identified compounds on taste may be further evaluated in human or animal taste tests.
1. An isolated nucleic acid encoding a G protein coupled receptor (GPCR) polypeptide involved in bitter taste transduction which polypeptide is at least 95% identical to the polypeptide of SEQ ID NO:4 or which encodes a polypeptide which is at least 95% identical to the entire extracellular or the transmembrane region of said GPCR polypeptide.
2. The isolated nucleic acid of claim 1 that encodes a polypeptide at least 96% identical to the polypeptide in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
3. The isolated nucleic acid of claim 1 that encodes a polypeptide at least 97% identical to the polypeptide in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
4. The isolated nucleic acid of claim 1 that encodes a polypeptide at least 96% identical to the polypeptide in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
5. The isolated nucleic acid of claim 1 that encodes a polypeptide at least 98% identical to the polypeptide in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
6. The isolated nucleic acid of claim 1 that encodes a polypeptide at least 99% identical to the polypeptide contained in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
7. The isolated nucleic acid of claim 1 that encodes a polypeptide identical to the polypeptide contained in SEQ ID NO:4 or to the entire extracellular or the transmembrane region of said GPCR polypeptide.
8. The isolated nucleic acid of claim 1 which comprises the nucleic acid sequence contained in SEQ ID NO:3 or the entire extracellular or the transmembrane coding region of said GPCR polypeptide.
9. The isolated nucleic acid of claim 1 which is contained in a plasmid.
10. An isolated or recombinant cell containing a nucleic acid according to claim 1 .
11. The isolated cell or recombinant cell of claim 10 which is selected from a bacterial, yeast, amphibian, mammalian and oocyte cell.
12. The isolated cell or recombinant cell of claim 10 which is a BHK, COS, HEK or CHO cell.
