MICRODEVICES FOR SCREENING BIOMOLECULES
Methods and devices for the parallel, in vitro screening of biomolecular activity using miniaturized microfabricated devices are provided. The biomolecules immobilized on the surface of the devices of the present invention include proteins, polypeptides, polynucleotides, polysaccharides, phospholipids, and related unnatural polymers of biological relevance. These devices are useful drug development, functional proteomics and clinical diagnostics and are preferably used for the parallel screening of families of related proteins.

47 . A device for analyzing components of a fluid sample, comprising: a substrate comprising a plurality of parallel microchannels comprising an array of at least 10 different proteins of a protein family.
48 . The device of claim 47 wherein the proteins are immobilized at a density of at least 100/cm 2 .
49 . The device of claim 47 , wherein the proteins are functionally related.
50 . The device of claim 47 , wherein the proteins are kinases.
51 . The device of claim 47 , wherein the proteins are recombinant proteins.
52 . The device of claim 47 , wherein the proteins are recombinant kinases.
53 . The device of claim 47 , wherein the proteins comprise antibodies.
54 . The device of claim 47 , wherein the proteins comprise capture agents.
55 . The device of claim 47 , wherein the proteins are functionally related or suspected of being functionally related.
56 . The device of claim 47 , wherein the proteins are immobilized at a density of at least 1000/cm 2 .

The device of claim 47 , wherein the protein family is selected from the group consisting of growth factor receptors, catecholamine receptors, amino acid derivative receptors, cytokine receptors, extracellular matrix receptors, immunoglobulins, lectins, cytokines, serpins, proteases, kinases, phosphatases, ras-like GTPases, hydrolases, steroid hormone receptors, heat-shock transcription factors, zinc-finger proteins, leucine-zipper proteins, homeodomain proteins, hepatitis C virus (HCV) proteases and HIV proteases.
59 . The device of claim 47 , wherein the parallel microchannels are separated by a sealing gasket.
60 . The device of claim 47 , wherein the device further comprises a cover above the microchannels.
61 . The device of claim 47 , wherein the proteins have a protein activity.
62 . The device of claim 61 , wherein the proteins are immobilized at a density of at least 1000/cm 2 .
63 . The device of claim 47 , wherein the array comprises at least 100 different proteins.
64 . The device of claim 47 , wherein the array comprises at least 1000 different proteins.

A device for analyzing components of a fluid sample comprising: a substrate comprising a plurality of parallel microchannels comprising an array of at least 100 different immobilized recombinant kinases, wherein the recombinant kinases are immobilized in the microchannels at a density of at least 100/cm 2 .
66 . The device of claim 65 , wherein the recombinant kinases each comprises an affinity tag.
67 . The device of claim 65 , wherein the immobilized kinases have kinase activity.
68 . The device of claim 65 , wherein the array comprises at least 1000 different recombinant kinases.
