Abstract

The present invention relates to a novel class of disubstituted aniline compounds. These compounds can inhibit histone deacetylase and are suitable for use in selectively inducing terminal differentiation, and arresting cell growth and/or apoptosis of neoplastic cells, thereby inhibiting proliferation of such cells. Thus, the compounds of the present invention are useful in treating a patient having a tumor characterized by proliferation of neoplastic cells. The compounds of the invention may also be useful in the prevention and treatment of TRX-mediated diseases, such as autoimmune, allergic and inflammatory diseases, and in the prevention and/or treatment of diseases of the central nervous system (CNS), such as neurodegenerative diseases. The present invention further provides pharmaceutical compositions comprising the compounds of the instant invention and safe dosing regimens of these pharmaceutical compositions, which are easy to follow, and which result in a therapeutically effective amount of these compounds in vivo.

Claims (43)

1. A compound of the following structural formula:

wherein

is optionally substituted aryl or optionally substituted heteroaryl;

is optionally substituted aryl or optionally substituted heteroaryl;

is aryl or heteroaryl;

R 1 is NH 2 , OH or NH[(C 1-6 alkyl)C(O)NR 5 R 6 ];

R 2 is H, Cl — 6 alkyl, O(C 1-6 alkyl), C(O)R 4 , (C 1-6 alkyl)C(O)NR 5 R 6 , C(O)(C 1-6 alkyl)NR 5 R 6 , (C 1-6 alkyl)NR 5 R 6 or SO 2 (C 1-6 alkyl);

R 3 is (C 1-6 alkyl)C(O)NR 5 R 6 , (C 1-6 alkyl)NR 5 R 6 ;

R 4 is H or C 1-6 alkyl;

R 5 is H, C 1-6 alkyl, aryl, heteroaryl or C(O)(C 1-6 alkyl), wherein said alkyl, aryl and heteroaryl groups are optionally substituted with one to three halo;

R 6 is H, C 1-6 alkyl, aryl, heteroaryl or C(O)(C 1-6 alkyl), wherein said alkyl, aryl and heteroaryl groups are optionally substituted with one to three halo;

or a stereoisomer or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 wherein

is aryl which is optionally substituted with one to three halo; or a stereoisomer or a pharmaceutically acceptable salt thereof.

3. The compound of claim 2 wherein

is heteroaryl; or a stereoisomer or a pharmaceutically acceptable salt thereof.

4. The compound of claim 3 wherein

is aryl or heteroaryl; or a stereoisomer or a pharmaceutically acceptable salt thereof.

5. The compound of claim 4 wherein

is phenyl, which is optionally substituted with one to three halo; or a stereoisomer or a pharmaceutically acceptable salt thereof.

6. The compound of claim 5 wherein

is thiophene; or a stereoisomer or a pharmaceutically acceptable salt thereof.

7. The compound of claim 6 wherein

is phenyl; or a stereoisomer or a pharmaceutically acceptable salt thereof.

8. A compound of claim 1 which is

N-[2-amino-5-(2-thienyl)phenyl]-4-[[2-(dimethylamino)ethyl](methyl sulfonyl)amino]benzamide;

N-[2-amino-5-(2-thienyl)phenyl]-4-[[2-(dimethylamino)-2-oxoethyl](methyl)amino]benzamide;

N-(4-aminobiphenyl-3-yl)-4-[bis(2-anilino-2-oxoethyl)amino]benzamide;

2,2′-{[4-({[2-amino-5-(3-thienyl)phenyl]amino carbonyl)phenyl]imino}diacetic acid;

N-[2-amino-5-(2-thienyl)phenyl]-4-{bis[2-(methylamino)-2-oxoethyl]amino}benzamide;

{[4-({[2-amino-5-(3-thienyl)phenyl]amino carbonyl)phenyl]amino}aceticacid;

N-[2-amino-5-(3-thienyl)phenyl]-4-{[2-(methylamino)-2-oxoethyl]amino}benzamide;

N-[2-amino-5-(3-thienyl)phenyl]-4-{[2-(benzylamino)-2-oxoethyl]aminobenzamide;

4-amino-N-[2-{[2-methylamino)-2-oxoethyl]amino}-5-(2-thienyl)phenyl]benzamide;

4-amino-N-[2-{[2-dimethylamino)-2-oxoethyl]amino}-5-(2-thienyl)phenyl]benzamide;

4-amino-N-[2-{[2-diethylamino)-2-oxo ethyl]amino 1-5-(2-thienyl)phenyl]benzamide;

4-({3-[acetyl (methyl)amino]propanoyl}amino)-N-[2-amino-5-(2-thienyl)phenyl]benzamide;

N-[2-amino-5-(2-thienyl)phenyl]-4-[methyl(4-methyl-5-oxohexanoyl)amino]benzamide;

4-(acetyl{2-[(4-chlorophenyl)amino]-1-methyl-2-oxoethyl}amino)-N-[2-amino-5-(2-thienyl)phenyl]benzamide;

N,N-dimethylglycyl-N 2 -(4-{[(2-amino-5-thien-2-ylphenyl)amino]carbonyl}phenyl)-N 1 -(4-chlorophenyl)alaninamide;

or a stereoisomer or a pharmaceutically acceptable salt thereof.

9. A pharmaceutical composition comprising a pharmaceutically effective amount of the compound according to claim 1 , and a pharmaceutically acceptable carrier.

10. A method of treatment of cancer in a mammal comprising the step of administering a therapeutically effective amount of the compound of claim 1 .
