Abstract

Disclosed is a process for making the compound of formula I: using the compounds of formulas II, Q, and XI or XII: wherein A is selected from the group consisting of Br, Cl and I (with Br being preferred); and R represents (C 1 -C 10 )alkyl. Also disclosed are intermediate compounds that are made in the disclosed process.

Claims (11)

1. A process for making a compound of Formula IV(i),

the process comprising:

(a) converting the compound of Formula II

to a compound of Formula IV:

wherein A, in Formulae II and IV in each instance is Br, CI or I; and

(b) hydrogenating the compound of Formula IV using a suitable hydrogenation catalyst under hydrogen pressure, to form the compound of Formula IV(i).

2. The process of claim 1 wherein substituent A is Br.

3. The process of claim 1 wherein step (a) comprises reacting said compound of Formula II with a suitable acid chloride in a suitable solvent at a suitable temperature, and then reacting the resulting reaction mixture with dimethylamine.

4. The process of claim 1 , wherein step (b) comprises making a mixture of said compound of Formula IV, a hydrogenation catalyst, and a suitable base, and pressurizing the mixture under H 2 pressure.

The process of claim 2 wherein, said conversion of the compound of Formula II in step (a) comprises: (i) reacting the compound of Formula II with an acid chloride which is thionyl chloride or oxalyl chloride at a suitable temperature in a solvent which is acetonitrile, THF, t-butylmethylether, methylenechloride, toluene, ethylacetate, diethylether, or chloroform; and (ii) reacting the reaction mixture from step (i) with dimethylamine wherein said dimethylamine is presented to the reaction mixture as dimethylamine gas or a solution of dimethylamine, and wherein, when dimethylamine is presented to the reaction mixture as a solution said solvent comprises acetonitrile, THF, t-butylmethylether, methylenechloride, toluene, ethylacetate, diethylether, or chloroform, and wherein said hydrogenation of the compound of Formula IV, step (b), comprises: (1) making a mixture of: said compound of Formula IV; a hydrogenation catalyst which is Pd/C, Pt/C, PdOH, or raney nickel; and a base which is KOH, NaOH, Na 2 CO 3 , K 2 CO 3 , NaHCO 3 , KHCO 3 , tetramethylguanidine, DBU, diisopropylethylamine or a mixture thereof; and (2) pressurizing the mixture under H 2 pressure.

The process of claim 5 wherein, in step 10 (a)(i), the acid chloride used is thionyl chloride and the solvent used to carry out the reaction of the compound of Formula II is acetonitrile, in step 10(b) dimethylamine is present in an amount of at least 1 mole equivalent with respect to said compound of Formula II, and in step 10(b), said hydrogenation catalyst used in step 10(b)(1) is Pd/C, and hydrogen is present at a pressure of from about 10 psi to about 500 psi.
