Diagnosis, prevention and treatment of crohn's disease using the OmpC antigen
View Patent ↗The present invention provides a method of diagnosing Crohn's disease in a subject by determining the presence or absence or IgA anti-OmpC antibodies in the subject, where the presence of the IgA anti-OmpC antibodies indicates that the subject has Crohn's disease.
1. A method for performing an assay for improving the diagnosis of Crohn's disease, said method comprising:
(a) contacting a patient's sample containing IgA anti-outer membrane protein antibodies (anti-OmpC antibodies) with an OmpC antigen, or reactive fragment thereof to form a complex between the OmpC antigen or reactive fragment thereof and the IgA anti-OmpC antibodies;
(b) contacting the sample from step (a) with an enzyme-labeled anti-IgA antibody to form a labeled complex;
(c) contacting the sample from step (b) containing a labeled complex with an enzyme substrate; and
(d) analyzing the sample from step (c) for the presence or absence of the labeled complex using a spectrophotometer, wherein the analysis detects the labeled complex, to improve the diagnosis of Crohn's disease.
2. The method of claim 1 , wherein said patient's sample is human serum.
3. The method of claim 1 , wherein said OmpC antigen comprises the amino acid sequence of SEQ ID NO: 1 or 90% identity thereof.
4. The method of claim 1 , wherein said assay is performed on an assay system, said assay system comprising:
(a) a plate device comprising an OmpC antigen, or reactive fragment thereof, wherein said OmpC antigen comprises the amino acid sequence of SEQ ID NO:1 or 90% identity thereof;
(b) an indicator secondary antibody specific for an OmpC autoantibody, wherein said indicator secondary antibody is an enzyme-labeled anti-IgA antibody; and
(c) instructions for detecting the presence or absence of a complex of said OmpC antigen, said OmpC autoantibody, and said indicator secondary antibody.
5. The method of claim 4 , wherein said plate device further comprises a control.
6. The method of claim 4 , wherein said system further comprises a substrate for said enzyme-labeled antibody.
7. The method of claim 6 , wherein said substrate is transformed to a product, wherein said product is chromogenic.
8. The method of claim 4 , wherein said OmpC antigen comprises the amino acid sequence of SEQ ID NO: 1.
9. The method of claim 1 , wherein said assay is performed to facilitate earlier and appropriate treatment for Crohn's Disease patients.
