Chimeric alphavirus replicon particles
Chimeric alphaviruses and alphavirus replicon particles are provided, including methods of making and using same. Specifically, alphavirus particles are provided having nucleic acid molecules derived from one or more alphaviruses and structural proteins (capsid and/or envelope) from at least two or more alphaviruses. Methods of making, using, and therapeutic preparations containing the chimeric alphavirus particle, are disclosed.
1. An alphavirus packaging cell line comprising one or more structural protein expression cassettes comprising sequences encoding one or more structural proteins, wherein at least one of said structural proteins is derived from two or more different alphaviruses, and wherein said one or more structural protein expression cassettes comprise cDNA copies of a defective helper RNA.
2. The packaging cell line of claim 1 , wherein said defective helper RNA directs expression of the structural protein(s).
3. The packaging cell line of claim 1 , wherein said defective helper RNA further comprises an alphavirus subgenomic promoter.
4. A method for producing recombinant alphavirus replicon particles, comprising introducing into the alphavirus packaging cell line of claim 1 an alphavirus replicon RNA derived from one or more alphaviruses, whereby an alphavirus particle comprising one or more heterologous RNA sequence(s) is produced.
5. The alphavirus packaging cell line of claim 1 wherein the structural protein derived from two or more different alphaviruses is a capsid protein.
6. The alphavirus packaging cell line of claim 1 wherein the structural protein derived from two or more different alphaviruses is an envelope glycoprotein.
7. An alphavirus packaging cell line comprising one or more structural protein expression cassettes comprising sequences encoding one or more structural proteins, wherein at least one of said structural proteins is derived from two or more different alphaviruses, and wherein one or more structural proteins comprise (a) a hybrid capsid protein having (i) an RNA binding domain derived from a first alphavirus and (ii) an envelope glycoprotein interaction domain derived from a second alphavirus which is different from the first alphavirus; and (b) an envelope glycoprotein derived from said second alphavirus.

The alphavirus packaging cell line of claim 1 wherein one or more structural proteins comprise an envelope glycoprotein having (i) a cytoplasmic tail portion derived from said first alphavirus and (ii) at least 95% of the remaining portion derived from a second alphavirus, wherein the second alphavirus is different from the first alphavirus.
9. The alphavirus packaging cell line of claim 1 wherein one of the two or more alphaviruses is Sindbis virus (SIN).

An alphavirus packaging cell line comprising one or more structural protein expression cassettes comprising sequences encoding one or more structural proteins, wherein at least one of said structural proteins is derived from two or more different alphaviruses, and wherein one of the two or more alphaviruses is Venezuelan equine encephalitis virus (VEE).
11. An alphavirus packaging cell line comprising one or more structural protein expression cassettes comprising sequences encoding one or more structural proteins, wherein at least one of said structural proteins is derived from two or more different alphaviruses, and wherein one of the two or more alphaviruses is SIN and the other of the two or more alphaviruses is VEE.
