Abstract

This invention provides methods of treating, suppressing, inhibiting, reducing an incidence, reducing the pathogenesis of, ameliorating the symptoms of, or ameliorating the secondary symptoms of a primary or recurring Herpes Simplex Virus (HSV) infection, or prolonging the latency to a relapse of an HSV infection, and disorders and symptoms associated with same and inducing an anti-HSV immune response in a subject comprising the step of contacting the subject with a composition comprising a mutant HSV strain comprising an inactivating mutation in a Us8 gene, followed by a second contacting with the composition.

Claims (16)

1. A method of inducing an anti-Herpes Simplex Virus (HSV) immune response in a subject comprising the steps of (a) administering a composition comprising an attenuated HSV mutant to said subject, wherein said attenuated HSV mutant consists of a single inactivating mutation, wherein said inactivating mutation is a deletion of base pairs (bp) 369-1479 of SEQ ID NO: 7, which is a Us8 gene, and (b) administering a booster dose of said composition comprising said attenuated HSV mutant, wherein said attenuated HSV mutant has a defect in neuronal spread in the anterograde and retrograde directions, wherein said booster dose enhances the immune response induced by the initial administration of said attenuated HSV mutant.

2. The method of claim 1 , wherein said booster dose is administered 3-6 weeks after said initial administration.

3. The method of claim 1 , wherein said attenuated mutant is deficient in cell-to-cell spread.

4. The method of claim 1 , wherein said attenuated mutant is replication-competent.

5. The method of claim 1 , wherein said anti-HSV immune response is an anti-HSV neutralizing antibody response.

6. The method of claim 1 , wherein said subject is infected by or is at risk for infection by HSV.

7. The method of claim 1 , wherein said initial administration is before exposure to HSV.

8. The method of claim 1 , wherein said initial administration is after exposure to HSV.

9. The method of claim 1 , wherein said subject is administered said composition at least one additional time after the administration of said booster dose.

10. The method of claim 1 , wherein said composition is administered intramuscularly, epidermally, subcutaneously, intravaginally, or via intra-respiratory mucosal injection.

A method of treating a Herpes Simplex Virus (HSV) infection in a subject comprising the step of (a) administering a composition comprising an attenuated HSV mutant to said subject, wherein said attenuated HSV mutant consists of a single inactivating mutation, wherein said inactivating mutation is a deletion of base pairs (bp) 369-1479 of SEQ ID NO: 7, which is a Us8 gene, and then (b) administering a booster dose of said composition comprising said attenuated HSV mutant, wherein said attenuated HSV mutant has a defect in neuronal spread in the anterograde and retrograde directions, wherein said booster dose enhances the immune response induced by the initial administration of said attenuated HSV mutant, wherein said booster dose is administered 3-6 weeks after said initial administration.

12. The method of claim 11 , wherein said HSV infection is an HSV-1 or HSV-2 infection.

13. The method of claim 11 , wherein said HSV infection is a primary HSV infection.

14. The method of claim 11 , wherein said HSV infection is HSV encephalitis, an HSV neonatal infection, or HSV labialis.

15. The method of claim 11 , wherein said HSV infection is a genital, oral, or ocular HSV infection.

16. The method of claim 11 , wherein said HSV infection is a flare, recurrence, or HSV labialis following a primary HSV infection.
