Abstract

This disclosure is directed, inter alia, to polynucleotides, polypeptides, vectors, cells and compositions comprising the same, and their use in affecting viral pathogenesis, in particular for influenza viral infection.

Claims (19)

1. An isolated polynucleotide comprising SEQ ID NO:8.

2. The isolated polynucleotide of claim 1 , further encoding a cytokine or growth factor.

3. A composition comprising the isolated polynucleotide of claim 1 .

4. An isolated polypeptide comprising SEQ ID NO:7.

5. The polypeptide of claim 4 , wherein said polypeptide is at least partially glycosylated.

6. The isolated polynucleotide of claim 1 , further comprising a vector.

7. The composition of claim 3 , further comprising a cytokine, a growth factor, an antiviral, an adjuvant, an excipient, a polynucleotide encoding a cytokine or growth factor, or a combination thereof.

8. An immunization method comprising:

administering a sufficient amount of an isolated polynucleotide comprising SEQ ID NO:8 to a subject;

wherein the subject is capable of resisting subsequent influenza viral challenge.

9. The method of claim 8 , wherein the amount of antibody titer specific to influenza hemagglutinin produced by the subject subsequent to viral challenge is increased by at least 2% as compared to a subject not administered a sufficient amount of the polynucleotide comprising SEQ ID NO:8.

10. The method of claim 8 , wherein the amount of interferon-γ produced by CD8-positive cells of the subject subsequent to viral challenge is increased by at least 2% as compared to a subject not administered a sufficient amount of the polynucleotide comprising SEQ ID NO:8.

11. The method of claim 8 , wherein the amount of chicken blood cell hemadsorption by challenge virus in solution with subject serum is reduced by at least 2% as compared to a subject not administered a sufficient amount of the polynucleotide comprising SEQ ID NO:8.

12. The method of claim 8 , wherein the post-challenge body temperature of the subject is maintained within 95% of the body temperature of the subject pre-challenge.

13. The method of claim 8 , wherein the post-challenge weight loss of a subject is at least 2% less as compared to a subject not administered a sufficient amount of the polynucleotide comprising SEQ ID NO:8.

14. The method of claim 8 , wherein the post-challenge survivability of a subject is improved as compared to a virally-challenged subject that was not administered an isolated polypeptide comprising SEQ ID NO:7.

15. The method of claim 8 , wherein the subsequent viral challenge is an influenza virus strain H5N1.

16. The method of claim 8 , wherein the subsequent viral challenge is an influenza viral strain that is the product of viral reassortment.

17. The method of claim 8 , wherein the subject is capable of resisting subsequent viral challenge with an influenza virus of an alternative clade or sub-clade than the initial viral challenge.
