Oxidative DNA damage protection
Provided herein are methods of screening compounds, gene sequences, and gene products in bacteria for agents that are protective against oxidative DNA damage in a human or animal. Gene sequences identified by these screens can also be used in diagnostic assays that identify subjects at increased risk for oxidative DNA damage. Pharmaceutical compositions that include DNA protective agents identified by these screens are also provided.
1. A method of treating or protecting against oxidative DNA damage in a subject, the method comprising:
(i) identifying a subject in need of treatment or protection against oxidative DNA damage; and
(ii) administering to the subject an effective amount of a polypeptide comprising amino acid residues 40-127 of PC4 (SEQ ID NO:2) with 8 or fewer amino acid substitutions.
2. The method of claim 1 , wherein the polypeptide
comprises amino acid residues 40-127 of PC4 (SEQ ID NO:2).
3. The method of claim 2 , wherein the polypeptide comprises amino acid residues 40-127 of SEQ ID NO:2 with 8 or fewer amino acid substitutions and does not comprise amino acid residues 1-39 of SEQ ID NO:2.
4. The method of claim 2 , wherein the polypeptide consists of amino acid residues 40-127 of SEQ ID NO:2.
5. The method of claim 1 , wherein the polypeptide sequence comprises SEQ ID NO:2.
6. The method of claim 1 , wherein the polypeptide consists of amino acid residues 40-127 of SEQ ID NO:2 with 8 or fewer amino acid substitutions.
7. The method of claim 1 , wherein the subject is a human.
