Abstract

The present disclosure provides compounds that function as modulators of aldehyde dehydrogenase (ALDH) enzymatic activity, as well as compositions and formulations comprising the compounds. The present disclosure provides therapeutic methods involving administering a subject compound, or a subject pharmaceutical composition.

Claims (64)

1. A compound of Formula I:

where X n and X y are each independently H, C, N, O, or a halogen; where n is the integer 0 or 1; where y is the integer 0 or 1;

where . . . (dotted line) is an optional bond; where z is the integer 0, 1, or 2;

where is an optional double bond;

wherein each of R 1 , R 2 , R 3 , R 4 , R 5 , R 6 , R 7 , R 8 , R 9 , and R 10 is independently selected from H; C; N; O; a halo; a substituted or unsubstituted phenyl group; an aliphatic group, an alkyl group; a substituted alkyl group; an alkenyl group; an alkynyl group; a substituted or unsubstituted cyclic group; a substituted or unsubstituted heterocyclic group; a substituted or unsubstituted aryl group; and a substituted or unsubstituted heteroaryl group;

wherein A is C or S; and where a=1 when A=C; and where a=2 when A=S;

wherein X b is C, N, O, or S; where b is the integer 0 or 1;

wherein X c is C, N, O, or S; where c is the integer 0 or 1;

wherein X d is C, N, O, or S; where d is the integer 0 or 1; and

wherein Ar is selected from a substituted aryl group, an unsubstituted aryl group, a substituted heteroaryl group, and an unsubstituted heteroaryl group, where aryl and heteroaryl are polycyclic;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

2. The compound of claim 1 , wherein Ar is a substituted naphthalene group.

3. A compound of Formula Ia:

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and . . . is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

wherein A=C or S, and where a=1 when A=C; and where a=2 when A=S;

wherein R 1 , R 2 , and R 3 are each independently selected from H; a halo; an aliphatic group, an alkyl group; a substituted alkyl group; a substituted or unsubstituted alkenyl group; and a substituted or unsubstituted alkynyl group;

wherein c is the integer 0 or 1;

wherein X d is C, N, O, or S; where d is the integer 0 or 1; and

wherein Ar is selected from a substituted aryl group, an unsubstituted aryl group, a substituted heteroaryl group, and an unsubstituted heteroaryl group, where aryl and heteroaryl are polycyclic;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

4. The compound of claim 1 , wherein the A compound has a structure selected from:

5. The compound of claim 1 , wherein the compound is of Formula Ib:

wherein X n and X y are each independently H, C, N, O, or a halogen; wherein n is the integer 0 or 1; where y is the integer 0 or 1;

wherein . . . (dotted line) is an optional bond;

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

wherein Ar is selected from a substituted aryl group, an unsubstituted aryl group, a substituted heteroaryl group, and an unsubstituted heteroaryl group, where aryl and heteroaryl are polycyclic;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

6. The compound of claim 1 , wherein the compound is of Formula Ic:

wherein X n and X y are each independently H, C, N, O, or a halogen; where n is the integer 0 or 1; where y is the integer 0 or 1;

wherein . . . (dotted line) is an optional bond;

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and . . . is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

where Ar is selected from a substituted aryl group, an unsubstituted aryl group, a substituted heteroaryl group, and an unsubstituted heteroaryl group, where aryl and heteroaryl are polycyclic;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

7. A compound of Formula Id:

wherein X n and X y are each independently H, C, N, O, or a halogen; where n is the integer 0 or 1; wherein y is the integer 0 or 1;

wherein . . . (dotted line) is an optional bond;

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and . . . is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

wherein A=C or S, and where a=1 when A=C; and where a=2 when A=S;

wherein X c is C, N, O, or S; where c is the integer 0 or 1; and

wherein R 1 and R 2 are each independently selected from H; a halo; an aliphatic group, an alkyl group; a substituted alkyl group; a substituted or unsubstituted alkenyl group; and a substituted or unsubstituted alkynyl group;

wherein R 3 is selected from a substituted polycyclic group, an unsubstituted polycyclic group, a substituted hetero polycyclic group, and an unsubstituted hetero polycyclic group;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

8. A compound of Formula Ie:

wherein X n and X y are each independently H, C, N, O, or a halogen; wherein n is the integer 0 or 1; where y is the integer 0 or 1;

wherein . . . (dotted line) is an optional bond;

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

wherein X b is C, N, O, or S; where b is the integer 0 or 1; and

wherein R 1 , R 2 , R 3 and R 4 are each independently selected from H; a halo; an aliphatic group; an alkyl group; a substituted alkyl group; a substituted or unsubstituted alkenyl group; and a substituted or unsubstituted alkynyl group;

wherein R 5 is selected from a substituted polycyclic group, an unsubstituted polycyclic group, a substituted hetero polycyclic group, and an unsubstituted hetero polycyclic group;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

9. A compound of Formula If:

wherein X n and X y are each independently H, C, N, O, or a halogen; wherein n is the integer 0 or 1; wherein y is the integer 0 or 1;

wherein . . . (dotted line) is an optional bond;

wherein z is the integer 0, 1, or 2, with the provisos that: 1) z=0 when X=halogen and . . . is not a bond; and 2) when z=0, X=O, . . . is not a bond, and one or more oxygen atoms (X) are present, oxygen is attached to a methyl group;

wherein is an optional double bond;

wherein A=C or S, and wherein a=1 when A=C; and wherein a=2 when A=S;

wherein R 1 , R 2 , R 3 , R 4 and R 5 are each independently selected from H; —OH; a halo; an aliphatic group; an alkyl group; a substituted alkyl group; a substituted or unsubstituted alkenyl group; and a substituted or unsubstituted alkynyl group;

wherein X c is C, N, O, or S; wherein c is the integer 0 or 1;

wherein X d is C, N, O, or S; wherein d is the integer 0 or 1; and

wherein Ar is selected from a substituted aryl group, an unsubstituted aryl group, a substituted heteroaryl group, and an unsubstituted heteroaryl group where aryl and heteroaryl are polycyclic;

or a pro-drug, a pharmaceutically acceptable salt, or a derivative thereof.

10. A pharmaceutical composition comprising:

a) a compound of claim 1 ; and

b) a pharmaceutically acceptable excipient.
