Abstract

The present invention provides for concentrated aqueous silk fibroin solutions and an all-aqueous mode for preparation of concentrated aqueous fibroin solutions that avoids the use of organic solvents, direct additives, or harsh chemicals. The invention further provides for the use of these solutions in production of materials, e.g., fibers, films, foams, meshes, scaffolds and hydrogels.

Claims (20)

1. A fiber produced by the method comprising processing a concentrated aqueous fibroin solution comprising a fibroin concentration of at least 10 wt % in order to form a fiber, wherein the fibroin solution is free of organic solvents and wherein the fibroin solution is produced by a process comprising dialyzing an aqueous silk fibroin solution against a hygroscopic polymer for a sufficient time to result in an aqueous fibroin solution of at least 10 wt %, wherein the hygroscopic polymer has a molecular weight of at least 8,000 g/mol.

2. The fiber of claim 1 , wherein the processed concentrated aqueous fibroin solution comprises dissolved silkworm silk, dissolved spider silk, or genetically engineered silk.

3. The fiber of claim 1 , further comprising a therapeutic agent.

4. The fiber of claim 3 , wherein the therapeutic agent is a small molecule, a protein, a peptide, or a nucleic acid.

The fiber of claim 3 , wherein the therapeutic agent is selected from the group consisting of antiinfectives, chemotherapeutic agents, anti-rejection agents, analgesics and analgesic combinations, anti-inflammatory agents, hormones, anti-angiogenic agents, anti-thrombotics, anti-metabolics, antimitotics, fibrinolytics, and growth factors.

The fiber of claim 5 , wherein the growth factor is selected from the group consisting of bone morphogenic proteins, bone morphogenic-like proteins, epidermal growth factor, fibroblast growth factor, platelet derived growth factor, insulin-like growth factor, transforming growth factors, and vascular endothelial growth factor.

7. The fiber of claim 3 , wherein the therapeutic agent is a small molecule selected from the group consisting of vitamins, sedatives, hypnotics, antibiotics, antiviral agents, steroids, growth factor inhibitors, growth factor promoters, prostaglandins and radiopharmaceuticals.

8. The fiber of claim 3 , wherein the therapeutic agent is selected from proteins, polysaccharides, glycoproteins, and lipoproteins.

9. The fiber of claim 3 , wherein the therapeutic agent is a protein.

10. The fiber of claim 3 , wherein the therapeutic agent is an antibody.

11. The fiber of claim 3 , wherein the therapeutic agent is present as a liquid or a finely divided solid.

12. The fiber of claim 3 , wherein the therapeutic agent is mixed with the aqueous silk fibroin solution.

13. The fiber of claim 3 , wherein the therapeutic agent is coated thereon.

14. The fiber of claim 3 , further comprising a pharmaceutically acceptable carrier.

15. The fiber of claim 3 , wherein the fiber is biodegradable.

16. The fiber of claim 1 , further comprising one or more additives.

17. The fiber of claim 16 , wherein the one or more additives is a biologically active compound.

18. The fiber of claim 17 , wherein the biologically active compound is selected from the group consisting of cell attachment mediators, biologically active ligands and substances that either promote or prevent cellular tissue ingrowth.

The fiber of claim 17 , wherein the biologically active compound is selected from the group consisting of collagen, elastin, fibronectin, vironectin, laminin, proteoglycans, Arg-Gly-Asp (RGD) peptides, bone morphogenic proteins, cytokines, epidermal growth factor, fibroblast growth factor, platelet-derived growth factor, insulin-like growth factor, and transforming growth factor.

20. The fiber of claim 16 , wherein the fiber is biodegradable.
