Abstract

The instant invention provides for compounds that inhibit the four known mammalian JAK kinases (JAK1, JAK2, JAK3 and TYK2) and PDK1. The invention also provides for compositions comprising such inhibitory compounds and methods of inhibiting the activity of JAK1, JAK2, JAK3 TYK2 and PDK1 by administering the compound to a patient in need of treatment for myeloproliferative disorders or cancer.

Claims (241)

1. A compound of the formula I:

wherein D is CR 3 ;

E is N or CR 3 ;

G is CR 3 ;

J is CR 3 ;

L is —NH —;

M is

(a) hydrogen,

(b) halo,

(c) hydroxyl,

(d) C 1-6 alkyl, optionally substituted with one to four substituents independently selected from the group consisting of halo, hydroxyl, SO 2 NR 4 R 5 , Si(CH 3 ) 3 and Si(CH 3 ) 3 O(C 1-6 haloalkyl),

(e) C 2-6 alkenyl,

(f) C 1-6 haloalkyl, which is optionally substituted with C 3-8 cycloalkyl,

(g) C 3-8 cycloalkyl,

(h) (C 1-6 alkyl)C 3-8 cycloalkyl,

(i) aryl, which is optionally substituted with one to four substituents independently selected from the group consisting of halo, hydroxyl, cyano, C 1-6 alkyl which is optionally substituted with one to three substituents independently selected from the group consisting of cyano, halo, hydroxyl and NR 4 R 5 , C 2-6 alkenyl which is optionally substituted with hydroxyl, C 2-6 alkynyl which is optionally substituted with hydroxyl or Si(CH 3 ) 3 , O(C 1-6 haloalkyl), C 3 - 8 cycloalkyl which is optionally substituted with hydroxyl, heteroaryl which is optionally substituted on either the carbon or heteroatom with R 8 , SO m NHR 7 , SO m R 7 , (C 1-6 alkyl)NHSO m R 7 , (C═O)R 8 , (C═O)OR 8 , (C═O)NHR 8 , (C═O)NH-C 1-3 alkyl-heterocyclyl, (C═NH)NHR 8 , (C═NOR 8 )C 1-3 alkyl, (C═NO-C 1-3 alkyl-(C═O)heterocyclyl)C 1-3 alkyl, and (C═NO-C 2-6 alkenyl)C 1-3 alkyl,

(j) heteroaryl, which optionally substituted on either the carbon or heteroatom with one to three substituents independently selected from the group consisting of halo, oxo, aryl and R 8 ;

(k) heterocyclyl,

(l) (C 1-6 alkyl)aryl, which is optionally substituted on either the alkyl or aryl group with a substituent selected from the group consisting of one to two halo and SO m NHR 7 ,

(m) C(O)R 8 , or

(n) NR 4 R 5 ;

R 1 is hydrogen;

R 2 is hydrogen;

R 3 is

(a) hydrogen,

(b) halo,

(c) hydroxyl,

(d) C 1-6 alkyl,

(e) C 2-6 alkenyl, optionally substituted with one to three hydroxyl,

(f) C 2-6 alkynyl, optionally substituted with one to two substituents independently selected from the group consisting of hydroxyl, heteroaryl and NR 4 R 5 ,

(g) C 1-6 haloalkyl,

(h) O(C 1-6 alkyl),

(i) O(C 1-6 haloalkyl),

(j) aryl,

(k) heteroaryl, optionally substituted on either the carbon or heteroatom with one to three substituents independently selected from the group consisting of hydroxyl, R 8 , C 1-6 alkyl(heterocyclyl), CH 2 (C═O)OR 8 and NR 4 R 5 ,

(l) heterocyclyl,

(m) B(OH) 2 ,

(n) NR 4 R 5 ,

(o) NHR 6 or

(p) NH(C═O)R 6 ;

R 4 is hydrogen or C 1-6 alkyl;

R 5 is hydrogen or C 1-6 alkyl;

R 6 is hydrogen, C 1-6 alkyl, NR 4 R 5 , —NH(C 1-6 alkyl)OR 8 , aryl, heteroaryl or heterocyclyl which is optionally substituted on either the carbon or heteroatom with R 8 , wherein said alkyl groups are optionally substituted with one to four substituents selected from hydroxy, halo, OR 8 , NR 8 SO 2 NR 4 R 5 , heteroaryl or heterocyclyl;

R 7 is hydrogen, C 1-6 alkyl, (C 3-6 cycloalkyl), heterocyclyl, (C 1-6 alkyl)heterocyclyl or (C 1-6 alkyl)heteroaryl, wherein said alkyl group is optionally substituted with one to four substituents selected from the group consisting of halo and hydroxy, and said heteroaryl group is optionally substituted with C 1-6 alkyl;

R 8 is hydrogen or C 1-6 alkyl, wherein said alkyl is optionally substituted with one to two hydroxyl; and,

m is an integer from zero to two;

or a pharmaceutically acceptable salt or stereoisomer thereof.

2. The compound of claim 1 , wherein E is N; or a pharmaceutically acceptable salt or stereoisomer thereof.

The compound of claim 2 , wherein M is C 1-6 alkyl, optionally substituted with one to four substituents independently selected from the group consisting of halo, hydroxyl and SO 2 NR 4 R 5 ; aryl, which is optionally substituted with one to four substituents independently selected from the group consisting of halo, hydroxyl, cyano, C 1-6 alkyl which is optionally substituted with one to three substituents independently selected from the group consisting of halo and hydroxyl, O(C 1-6 haloalkyl), C 3-8 cycloalkyl which is optionally substituted with hydroxyl, heteroaryl and SO m NHR 7 ; or heteroaryl, which is optionally substituted with halo; or a pharmaceutically acceptable salt or stereoisomer thereof.

The compound of claim 3 , wherein M is aryl, which is optionally substituted with one to four substituents independently selected from the group consisting of halo, hydroxyl, cyano, C 1-6 alkyl which is optionally substituted with one to three substituents independently selected from the group consisting of halo and hydroxyl, O(C 1-6 haloalkyl), C 3-8 cycloalkyl which is optionally substituted with hydroxyl, heteroaryl and SO m NHR 7 , or a pharmaceutically acceptable salt or stereoisomer thereof.

The compound of claim 2 , wherein R 3 is hydrogen, halo, C 2-6 alkenyl optionally substituted with one to three hydroxyl, C 2-6 alkynyl optionally substituted with one to two substituents independently selected from the group consisting of hydroxyl, heteroaryl and NR 4 R 5 , heteroaryl optionally substituted on either the carbon or heteroatom with one to three substituents independently selected from the group consisting of hydroxyl, R 8 , C 1-6 alkyl(heterocyclyl), CH 2 (C═O)OR 8 and NR 4 R 5 , or B(OH) 2 ; or a pharmaceutically acceptable salt or stereoisomer thereof.

6. The compound of claim 1 selected from:

5-(Cyclopropylamino)-9-fluorobenzo[c]-2,6-naphthyridin-1(2H)-one;

9-Fluoro-5-{[(1S)-2-methyl-1-(trifluoromethyl)propyl]amino}benzo[c]-2,6-naphthyridin-1(2H)-one;

9-Bromo-5-[(2,4,6-trifluorophenyl)amino]benzo[c]-2,6-naphthyridin-1(2H)-one;
