Polynucleotide primers
A polynucleotide comprising at least the final six nucleotides of one of the following primer sequences, or a sequence complementary thereto: SEQ. ID NOS. 3 to 16, 18, 20 to 33, 35 or 37 to 39. A method of detecting the presence or absence of a mutation in the PIK3CA gene, wherein the mutation is one of H1047R, H1047L, E542K and E545K, and preferably ARMS primers are combined with Scorpion primers.
1. A synthetic polynucleotide having the sequence of SEQ ID NO: 5.
2. The synthetic polynucleotide according to claim 1 further comprising a quencher group and a fluorophore group.
3. The synthetic polynucleotide according to claim 2 , wherein the quencher group and the fluorophore group are separated by a nucleotide tail sequence comprising first and second regions, the nucleotides of the first region being complementary to but in reverse order from the nucleotides of the second region, such that hybridization of the first region to the second group results in the quencher group to be sufficiently close to the fluorophore group to quench the fluorophore group.
4. The synthetic polynucleotide according to claim 3 , wherein the tail sequence further comprises a third region having a sequence complementary to a region of the PIK3CA gene.
5. The synthetic polynucleotide according to claim 2 , wherein the quencher group comprises Dabcyl.
6. The synthetic polynucleotide according to claim 2 , wherein the fluorophore comprises Hex, Fam or Rox,
7. A kit comprising at least one synthetic primer pair consisting of a forward primer having the sequence of SEQ ID NO: 5 and a reverse primer having the sequence of SEQ ID NO: 18.
8. The kit according to claim 7 , wherein the reverse primer comprises a quencher group and a fluorophore group.
9. The kit according to claim 8 , wherein the quencher group and the fluorophore group are separated by a nucleotide tail sequence comprising first and second regions, wherein nucleotides of the first region are complementary to, but in reverse order from, nucleotides of the second region, such that hybridisation of the first region to the second group results in the quencher group to be sufficiently close to the fluorophore group to quench the fluorophore group.
10. The kit according to claim 9 , wherein the tail sequence further comprises a third region having a sequence complementary to a region of the PIK3CA gene.
11. The kit according to claim 9 , wherein the tail sequence comprises SEQ. ID NO. 17.
