Abstract

Innovative graft polymers designed for the efficient delivery of antisense molecules into biological cells and for maintaining the biological activity of these molecules while in serum and other aqueous environments are provided. Such polymers may comprise an anionic graft polymer comprising an anionic polymer backbone with pendant carboxylic acid groups and pendant chains comprising amphipathic or hydrophilic polymers covalently bonded to a portion of said pendant carboxylic acid groups. Antisense molecule delivery vectors comprising such polymers in combination with cationic agents for delivery of antisense molecules are also disclosed.

Claims (16)

1. A biocompatible delivery vector comprising:

an anionic graft polymer comprising an anionic polymer backbone with pendent carboxylic acid groups and pendent chains comprising amphipathic polyetheramines propylene oxide moieties, bonded to a portion of said pendent carboxylic acid groups, wherein (a) the anionic graft polymer is physically bound to a complex comprising at least one cationic agent physically bound to at least one anionic antisense molecule, (b) the absolute value of the positive-to-negative (+/−) charge ratio of the delivery vector ranges from about 0.25 to about 4.7 between said anionic graft polymer and said complex of said cationic agent and said antisense molecule, (c) the molecular weight of the covalently bonded amphipathic polyetheramine moieties are between about 200 Daltons and about 5,000 Daltons; and (d) the anionic polymer has a graft density between about 1 and about 25 mole percent.

2. The delivery vector of claim 1 , wherein the at least one cationic agent is selected from a group consisting of surfactants, liposomes, peptides, polymers, micelles, nanoparticles and combinations thereof.

The delivery vector of claim 1 , wherein said anionic backbone polymer is a polyanhydride, poly(acrylic acid), poly(alkylacrylic acid), carboxymethylcellulose, poly acid, polyaspartic acid, or vinyl copolymers and said pendent chains comprise poly(ether-amines), polyalkylene oxides, polyolefinic alcohols, polyvinyl amines, or combinations thereof.

4. The delivery vector of claim 1 , wherein said anionic backbone polymer is a poly(propyl-acrylic acid), said pendent chains predominantly comprise ethylene oxide repeating units, and said polymer has a graft density between about 5 and about 25 mole percent.

5. The delivery vector of claim 1 having an absolute value of the positive-to-negative (+/−) charge ratio of about 1 between said anionic graft polymer, said cationic agent and said antisense molecule.

6. The delivery vector of claim 1 having a size of about 25 nm to about 300 nm.

7. The delivery vector of claim 1 , wherein the antisense molecule is an antisense oligonucleotides.

8. The delivery vector of claim 1 , wherein the antisense molecule is a siRNA.

9. A pharmaceutical composition comprising the delivery vector of claim 1 , and a pharmaceutically acceptable carrier.

10. A method of silencing oncogenic expression in cancer cells, the method comprising:

administering to a patient a therapeutically effective amount of the delivery vector of claim 1 , further comprising an siRNA targeted to an anti-apoptotic gene.

11. The method of claim 10 , wherein the sIRNA concentration is between about 40 and about 50 nM.

12. The method of claim 10 , wherein the anti-apoptotic gene is BcL-2.

13. The method of claim 10 , wherein the charge ratio between said anionic graft polymer, said cationic agent and said antisense molecule is about 1.

14. The method of claim 10 , wherein the delivery vector has a size of about 25 nm to about 300 nm.
