Abstract

Methods for enhancing or stimulating hematopoiesis including the step of administering Interleukin-12 (IL-12) to yield hematopoietic recovery in a mammal in need. Preferred methods include the step of administering IL-12 as an adjuvant therapy to alleviate the hematopoietic toxicities associated with one or more treatment regimens used to combat a disease state. Other methods include administering IL-12 to ameliorate various hematopoietic deficiencies. Still other methods are directed to uses of IL-12 for in-vivo proliferation of hematopoietic repopulating cells, hematopoietic progenitor cells and hematopoietic stem cells. Other disclosed methods are directed to uses of IL-12 for bone marrow preservation or recovery.

Claims (36)

1. A method for bone marrow preservation or recovery in a mammal having a hematopoietic malignancy comprising:

(a) administering a treatment to a mammal having a hematopoietic malignancy, wherein the mammal does not have a solid tumor and the treatment results in the near destruction of bone marrow in the mammal; and

(b) administering a single therapeutically effective dose of IL-12 to the mammal, without the use of hematopoietic repopulating cells, hematopoietic progenitor cells or hematopoietic stem cells, wherein administration of the single therapeutically effective dose of IL-12 results in bone marrow preservation or recovery,

wherein step (b) is performed before, during or after step (a); and

wherein the dose of IL-12 is less than 500 ng/kg.

2. The method of claim 1 , wherein the mammal has bone marrow failure.

3. The method of claim 1 , wherein the administration of the single therapeutically effective dose of IL-12 obviates the need for a bone marrow transplant.

4. The method of claim 1 , wherein bone marrow preservation or recovery includes an increase in hematopoietic repopulating cells, hematopoietic stem cells or hematopoietic progenitor cells.

5. The method of claim 1 , wherein bone marrow preservation or recovery includes an increase in one or more differentiated hematopoietic cells types.

6. The method of claim 1 , wherein bone marrow preservation or recovery includes an increase in hematopoietic support cells.

7. The method of claim 1 , wherein the mammal has a deficiency in one or more hematopoietic cell types or lineages.

8. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising lymphopenia.

9. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising myelopenia.

10. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising leukopenia.

11. The method of claim 10 , wherein the leukopenia is neutropenia.

12. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a erythropenia.

13. The method of claim 1 , wherein the hematopoietic malignancy results in a deficiency comprising megakaryopenia.

14. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in platelets.

15. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in monocytes.

16. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in lymphocytes.

17. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in erythrocytes.

18. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in neutrophils.

19. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in T cells.

20. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in granulocytes.

21. The method of claim 1 , wherein the hematopoietic malignancy results in a hematopoietic deficiency comprising a deficiency in dendritic cells.

22. The method of claim 1 , wherein the dose of IL-12 is less than 200 ng/kg.

23. The method of claim 1 , wherein the dose of IL-12 is less than 100 ng/kg.

24. The method of claim 1 , wherein the dose of IL-12 is less than 10 ng/kg.

25. The method of claim 1 , wherein the hematological malignancy is a hyperproliferative stem cell disorder.

26. The method of claim 1 , wherein the treatment is radiation therapy.

27. The method of claim 1 , wherein the treatment is chemotherapy.

28. The method of claim 1 , wherein the treatment is a combination of radiation therapy and chemotherapy.

29. The method of claim 26 , wherein the treatment comprises a high dose treatment modality.

30. The method of claim 26 , wherein the treatment comprises a dose dense treatment modality.

31. The method of claim 27 , wherein the treatment comprises a high dose treatment modality.

32. The method of claim 27 , wherein the treatment comprises a dose dense treatment modality.
