Abstract

The present invention discloses an attenuated recombinant alphavirus that is incapable of replicating in mosquito cells and of transmission by mosquito vectors. These attenuated alphavirus may include but is not limited to Western Equine Encephalitis virus, Eastern equine encephalitis virus, Venezuelan equine encephalitis virus or Chikungunya virus. The present invention also discloses the method of generating such alphaviruses and their use as immunogenic compositions.

Claims (15)

1. An attenuated recombinant alphavirus comprising: an alphavirus nucleic acid, having (i) an insertion of an internal ribosomal entry site of encephalomyelocarditis virus (EMCV IRES) between the end of nonstructural protein 4 (nsP4) coding sequence and initiating AUG of a subgenomic RNA coding sequence of the alphavirus, and (ii) an inactivated subgenomic promoter, wherein the alphavirus is attenuated.

2. The attenuated alphavirus of claim 1 , wherein the 5′ UTR of the subgenomic RNA between the end of nonstructural protein 4 (nsP4) coding sequence and the initiating AUG of a subgenomic RNA coding sequence of the alphavirus nucleic acid is deleted.

The attenuated alphavirus of claim 1 , further comprising clustered point mutations in the alphavirus nucleic acid located in the 5′ UTR of the subgenomic RNA that inactivate the alphavirus subgenomic promoter, the mutations comprising two or more mutations at position 5, 8, 11, 12, 14, 17, 20, 22, 24, 25, 26, 27, 28, 29, 30, 32, 33, 34, 35, 36, 37 or 38 of SEQ ID NO:1.

4. The attenuated alphavirus of claim 3 , wherein the mutation of the 5′ UTR of the subgenomic RNA does not modify the amino acid sequence of the carboxy terminus of nsP4.

The attenuated alphavirus of claim 1 , further comprising adaptive mutations in the nucleic acid sequence encoding the non-structural protein 2 (nsP2) resulting in an amino acid change corresponding to a Y370C mutation in TC-83 virus that results in an increase in virus replication, release, and titer.

6. The attenuated alphavirus of claim 1 , wherein the alphavirus is Chikungunya virus.

7. The attenuated alphavirus of claim 1 , wherein the alphavirus is Eastern Equine Encephalitis Virus.

8. The attenuated alphavirus of claim 1 , wherein the alphavirus is Venezuelan Equine Encephalitis virus.

9. The attenuated alphavirus of claim 1 , wherein the alphavirus is Western Equine Encephalitis virus.

10. The attenuated recombinant alphavirus of claim 1 , wherein the alphavirus is incapable of replicating in mosquitoes and mosquito cells.

An expression vector comprising an alphavirus nucleic acid having (i) an insertion of an internal ribosomal entry site of encephalomyelocarditis virus (EMCV IRES) between the end of nonstructural protein 4 (nsP4) coding sequence and initiating AUG of a subgenomic RNA coding sequence of the alphavirus, and (ii) an inactivated subgenomic promoter.

12. An isolated host cell comprising the expression vector of claim 11 .

13. A pharmaceutical composition comprising the attenuated recombinant alphavirus of claim 1 and a pharmaceutically acceptable carrier.

14. A method for inducing an anti-alphavirus immune response in a subject, comprising administering to the subject an attenuated recombinant alphavirus of claim 1 .

15. The method of claim 14 , wherein the subject is a human, a horse or other domestic animal.
