Stable cell lines and methods for evaluating gastrointestinal absorption of chemicals
Nucleic acids and vectors for interfering with the expression of membrane efflux transport proteins in cells that express such proteins are provided. Also provided are cells and cell lines comprising such nucleic acids and vectors. Methods for screening chemicals and biomolecules for gastrointestinal absorption in animals, and kits for practicing such methods are also provided.
1. A host cell transformed with a vector comprising a nucleic acid sequence encoding a nucleic acid molecule for inhibiting expression of p-glycoprotein, wherein the nucleic acid sequence comprises SEQ ID NO: 1, 2, 3, 4, or 5.
2. The host cell of claim 1 , wherein the cell expresses p-glycoprotein.
3. The host cell of claim 1 , wherein the cell is an intestinal epithelial cell.
4. A cell culture comprising the host cell of claim 3 .
5. The host cell of claim 1 , wherein said host cell has been deposited with the American Type Culture Collection and assigned accession number PTA-8752 or PTA-8753.
6. The host cell of claim 1 , wherein the cell is a Caco-2 cell.
7. The host cell of claim 1 , wherein the cell is derived from a Caco-2 cell.
8. The host cell of claim 1 , wherein the cell is a C2BBe1 cell.
9. A kit for determining the absorption characteristics of a chemical compound, comprising the cell of claim 1 .
10. A nucleic acid sequence comprising SEQ ID NO: 1, 2, 3, 4, or 5.
11. A nucleic acid encoded by the nucleic acid sequence of claim 10 .
12. A vector comprising a nucleic acid sequence encoding a nucleic acid molecule for inhibiting expression of p-glycoprotein, wherein the nucleic acid sequence comprises SEQ ID NO: 1, 2, 3, 4, or 5.
