Abstract

The invention discloses immunogenic polypeptides comprising several MAGE-specific antigen epitopes selected from different (i.e. discrete) members of the MAGE protein family, nucleic acids coding therefor, recombinant viruses and/or cells comprising said nucleic acids, and compositions thereof. Methods for eliciting or inducing MAGE-specific immune responses utilizing the aforementioned immunogenic agents are also disclosed.

Claims (17)

1. A method of inducing an immune response against a MAGE protein in an animal comprising administering to the animal a nucleic acid molecule encoding a polypeptide comprising a first amino acid sequence Glu Ser Leu Gln Leu Val Phe Gly Ile Asp Val Lys Glu Ala Asp Pro Thr Gly His Ser Tyr Val Leu Val Thr Cys Leu Gly Leu Ser Tyr Asp Gly (amino acid residues 2-34 of SEQ ID NO: 1) and a second amino acid sequence Glu Val Asp Pro Ile Gly His Leu Tyr (SEQ ID NO: 6).

2. The method of claim 1 wherein the first and second amino acid sequences are separated by a protease cleavage site.

3. The method of claim 2 wherein the protease cleavage site has the amino acid sequence Asn Lys Arg Lys (amino acid residues 35-38 of SEQ ID NO: 1).

4. The method of claim 1 wherein the first amino acid sequence is Met Glu Ser Leu Gln Leu Val Phe Gly Ile Asp Val Lys Glu Ala Asp Pro Thr Gly His Ser Tyr Val Leu Val Thr Cys Leu Gly Leu Ser Tyr Asp Gly (amino acid residues 1-34 of SEQ ID NO: 1).

5. The method of claim 4 wherein the protease cleavage site has the amino acid sequence Asn Lys Arg Lys (amino acid residues 35-38 of SEQ ID NO: 1).

6. The method of claim 1 wherein the nucleic acid molecule Comprises SEQ ID NO: 2.

7. The method of claim 1 wherein the polypeptide comprises SEQ ID NO: 1.

A method of inducing an immune response against a MAGE protein in an animal comprising administering to the animal a recombinant expression vector comprising a nucleic acid molecule encoding a polypeptide comprising a first amino acid sequence Glu Ser Leu Gln Leu Val Phe Gly Ile Asp Val Lys Glu Ala Asp Pro Thr Gly His Ser Tyr Val Leu Val Thr Cys Leu Gly Leu Ser Tyr Asp Gly (amino acid residues 2-34 of SEQ ID NO: 1) and a second amino acid sequence Glu Val Asp Pro Ile Gly His Leu Tyr (SEQ ID NO: 6).

9. The method of claim 8 wherein the expression vector is selected from the group consisting of a, plasmid, and a viral vector.

10. The method of claim 9 wherein the viral vector is selected from the group consisting of adenovirus, alphavirus and poxvirus.

11. The method of claim 10 wherein the poxvirus is ALVAC or ALVAC(2).

12. The method of claim 1 wherein the nucleic acid molecule is administered as part of a composition comprising a pharmaceutically acceptable carrier.

13. The method of claim 8 wherein the recombinant expression vector is administered as part of a composition comprising a pharmaceutically acceptable carrier.

14. The method of claim 1 wherein the animal is a human being.

15. The method of claim 8 wherein the animal is a human being.

16. The method of claim 12 wherein the animal is a human being.

17. The method of claim 13 wherein the animal is a human being.
