Abstract

Pyrimidone derivatives of formula (I) wherein R1, R2, R3, R4, R5, and R6 are as defined in the disclosure. Also disclosed are methods of preparing the compounds of formula (I) and their use in therapeutics.

Claims (42)

1. A compound of formula (I):

wherein:

R1 represents a 4-pyridine ring;

R2 represents a hydrogen atom;

R3 represents a hydrogen atom;

R4 represents:

a hydrogen atom, or

a phenyl ring which is optionally substituted by 1 to 4 substituents selected from a C 1-6 alkyl group, a halogen atom, a C 1-2 halogenated alkyl group, a hydroxyl group, a C 1-6 alkoxy group, and a C 1-2 halogenated alkoxy group;

R5 represents a hydrogen atom, a C 1-6 alkyl group, a 4-pyridine group, or a phenyl group, these groups being optionally substituted by 1 to 4 substituents selected from a C 1-6 alkyl group, a halogen atom, a C 1-2 halogenated alkyl group, a hydroxyl group, a C 1-6 alkoxy group, and a C 1-2 halogenated alkoxy group;

R6 represents a hydrogen atom, a C 1-6 alkyl group, a benzyloxy group, or a phenyl-C 1-6 alkyl group, these groups being optionally substituted by 1 to 4 substituents selected from a C 1-6 alkyl group, a halogen atom, a C 1-2 halogenated alkyl group, a hydroxyl group, a C 1-6 alkoxy group, and a C 1-2 halogenated alkoxy group;

or an acid addition salt thereof;

with the proviso that when R4 and R5 represent a hydrogen atom, R6 is not methyl or benzyl.

2. The compound of formula (I) according to claim 1 :

wherein

R1 represents a 4-pyridine ring;

R2 represents a hydrogen atom;

R3 represents a hydrogen atom;

R4 represents a hydrogen atom, a halogen atom, or a phenyl group;

R5 represents a hydrogen atom, a C 1-6 alkyl group, a 4-pyridine ring, or a phenyl group;

R6 represents a hydrogen atom, a C 1-6 alkyl group, a benzyloxy group, or a phenyl C 1-6 alkyl group, these groups being optionally substituted by 1 to 4 substituents selected from a C 1-6 alkyl group, a halogen, and a C 1-6 alkoxy group;

or an acid addition salt thereof;

with the proviso that when R4 and R5 represent a hydrogen atom, R6 is not methyl or benzyl.

3. The compound according to claim 1 , selected from the group consisting of:

2-Pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

7-Phenyl-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

8,9-Dimethyl-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

8-Phenyl-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

2,8-Di-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

9-Benzyloxy-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

9-[2-(2-Methoxy-phenyl)-ethyl]-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

2-Pyridin-4-yl-9-(2-o-tolyl-ethyl)-pyrido[1,2-a]pyrimidin-4-one;

9-(3-Phenyl-propyl)-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

9-[2-(2,4-Difluoro-phenyl)-ethyl]-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

9-[2-(2-Fluoro-phenyl)-ethyl]-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one; and

9-[3-(2-Fluoro-phenyl)-propyl]-2-pyridin-4-yl-pyrido[1,2-a]pyrimidin-4-one;

or an acid addition salt thereof.

4. A pharmaceutical composition comprising a compound of formula (I) according to claim 1 or a pharmaceutically acceptable salt, hydrate or solvate thereof and one or more pharmaceutical additives.

5. A pharmaceutical composition comprising a compound of formula (I) according to claim 2 or a pharmaceutically acceptable salt, hydrate or solvate thereof and one or more pharmaceutical additives.

6. A pharmaceutical composition comprising a compound according to claim 3 or a pharmaceutically acceptable salt, hydrate or solvate thereof and one or more pharmaceutical additives.

7. A method of inhibiting the activity of glycogen synthase kinase 3-beta (GSK3-β), which comprises administering to a patient in need of said inhibition a therapeutically effective amount of a compound of formula (I) according to claim 1 or a pharmaceutically acceptable salt thereof.

8. A method of treating malaria in a patient, comprising administering to said patient a therapeutically effective amount of a compound of formula (I) according to claim 1 or a pharmaceutically acceptable salt thereof.

9. A method of treating Pemphigus vulgaris in a patient, comprising administering to said patient a therapeutically effective amount of a compound of formula (I) according to claim 1 or a pharmaceutically acceptable salt thereof.
