Abstract

RNA aptamers and methods for identifying the same are disclosed. The RNA aptamers selectively bind coagulation factors, E2F family members, Ang1 or Ang2, and therapeutic and other uses for the RNA aptamers are also disclosed.

Claims (20)

1. An RNA aptamer that binds to the Tie2 ligand Ang2, wherein the aptamer comprises a sequence which is at least 80% identical to a nucleotide sequence selected from the group consisting of SEQ ID NOs:168-187, or a truncate thereof.

2. The aptamer of claim 1 , wherein the aptamer comprises a least one stem structure and at least one loop structure, and wherein the aptamer further comprises a consensus sequence consisting of AUCAGC.

3. The aptamer of claim 1 , comprising a nucleotide sequence selected from the group consisting of SEQ ID NOs:168-187, or a truncate thereof.

4. The aptamer of claim 1 , having a dissociation constant of about 100 picomolar (pM) to about 10 nanomolar (nM) or less.

5. The aptamer of claim 1 , wherein the aptamer comprises at least one 2′-modified nucleotide.

6. The aptamer of claim 5 , wherein the 2′-modified nucleotide is selected from the group consisting of a 2′-halo-modified nucleotide, a 2′-fluoro-modified nucleotide, and a 2′-O-alkyl-modified nucleotide.

7. The aptamer of claim 5 , comprising at least one 2′-fluorocitidine and/or one 2′-fluorouridine.

8. The aptamer of claim 5 , wherein all cytidines are 2′-deoxy-2′-fluorocytidine, and/or all uridines are 2′-deoxy-2′-fluorouridine.

9. The aptamer of claim 5 , comprising at least one 2′-O-alkyl-modified nucleotide.

10. The aptamer of claim 1 , wherein the aptamer consists of about 15 to 100 bases.

11. The aptamer of claim 1 , wherein the aptamer further comprises a covalently linked carrier selected from the group consisting of a soluble polymer, a biodegradable polymer, polyethylene glycol, and cholesterol.

A pharmaceutical composition comprising a therapeutically effective amount of an RNA aptamer which binds to the Tie2 ligand Ang2, wherein the aptamer comprises a sequence which is at least 80% identical to a nucleotide sequence selected from the group consisting of SEQ ID NOs:168-187, or atruncate thereof.

13. The composition of claim 12 , wherein the aptamer comprises a least one stem structure and at least one loop structure, and wherein the aptamer further comprises a consensus sequence consisting of AUCAGC.

14. The composition of claim 12 , wherein the aptamer comprises a nucleotide sequence selected from the group consisting of SEQ ID NOs:168-187.

15. The composition of claim 12 , wherein the aptamer comprises at least one 2′-modified nucleotide.

16. The composition of claim 15 , wherein the aptamer comprises at least one modified nucleotide selected from the group consisting of a 2′-halo-modified nucleotide, a 2′-fluoro-modified nucleotide, and a 2′-O-alkyl-modified nucleotide.

17. The composition of claim 15 , wherein the aptamer comprises cytidines that are all 2′-deoxy-2′-fluorocytidine, and/or uridines that are all 2′-deoxy-2′-fluorouridine.

18. The composition of claim 12 , wherein the aptamer further comprises a covalently linked carrier selected from the group comprising; a soluble polymer, a biodegradable polymer, polyethylene glycol, or cholesterol.

19. The aptamer of claim 1 , wherein the aptamer comprises a least one stem structure and at least one loop structure, and wherein the aptamer further comprises a consensus sequence consisting of CAGCUC.

20. The composition of claim 12 , wherein the aptamer comprises a least one stem structure and at least one loop structure, and wherein the aptamer further comprises a consensus sequence consisting of CAGCUC.
