Inhibitors of Janus kinases
View Patent ↗The instant invention provides for compounds that inhibit the four known mammalian JAK kinases (JAK1, JAK2, JAK3 and TYK2) and PDK1. The invention also provides for compositions comprising such inhibitory compounds and methods of inhibiting the activity of JAK1, JAK2, JAK3, TYK2 and PDK1 by administering the compound to a patient in need of treatment for myeloproliferative disorders or cancer.
1. A compound of formula I:
wherein
L is C;
M is C;
Q is CHR 3 ;
U is NR 5 ;
V is C═O;
the dashed circle which represents optional aromatic bonds is absent;
R 1 is aryl or heteroaryl, wherein said aryl and heteroaryl groups are optionally substituted on either the carbon or the heteroatom with one to three substituents independently selected from the group consisting of halo, hydroxyl, cyano, C 1-6 alkyl and heterocyclyl, wherein said alkyl group is optionally substituted with hydroxyl or cyano, and said heterocyclyl group is optionally substituted with halo;
R 3 is hydrogen;
R 5 is hydrogen, halo, cyano, C 1-6 alkyl, (C 1-6 alkyl)OR 6 , heteroyclyl, CH 2 -heterocyclyl or CH 2 -heteroaryl, wherein said alkyl groups are optionally substituted with one to three substituents independently selected from the group consisting of halo, hydroxyl and NR 6 R 7 , and said heterocyclyl groups are optionally substituted on either the carbon or the heteroatom with C 1-6 alkyl;
R 6 is hydrogen or C 1-6 alkyl, wherein said alkyl group is optionally substituted with hydroxyl or C 3-6 cycloalkyl;
R 7 is hydrogen or C 1-6 alkyl, wherein said alkyl group is optionally substituted with hydroxyl;
or a pharmaceutically acceptable salt thereof.

The compound of claim 1 wherein R 1 is aryl, wherein said aryl group is substituted with one to three substituents independently selected from the group consisting of halo and C 1-6 alkyl, wherein said alkyl group is optionally substituted with hydroxyl; or a pharmaceutically acceptable salt thereof.
3. The compound of claim 2 wherein R 1 is phenyl, wherein said phenyl group is substituted with one to three substituents independently selected from the group consisting of halo and C 1-6 alkyl, wherein said alkyl group is optionally substituted with hydroxyl; or a pharmaceutically acceptable salt thereof.
4. The compound of claim 3 wherein
R 5 is C 1-6 alkyl, wherein said alkyl group is optionally substituted with one to three hydroxyl; or a pharmaceutically acceptable salt thereof.
