Pyridyl amide T-type calcium channel antagonists
View Patent ↗The present invention is directed to pyridyl amide compounds which are antagonists of T-type calcium channels, and which are useful in the treatment or prevention of disorders and diseases in which T-type calcium channels are involved. The invention is also directed to pharmaceutical compositions comprising these compounds and the use of these compounds and compositions in the prevention or treatment of such diseases in which T-type calcium channels are involved.
1. A method for the treatment of schizophrenia which comprises administering to a human patient in need of such treatment an effective amount of a compound of the formula:
wherein:
R 1a is selected from the group consisting of:
(1) halogen,
(2) phenyl, which is unsubstituted or substituted with halogen, hydroxyl, C 1-6 alkyl, —O—C 1-6 alkyl, C 3-6 cycloalkyl, or —NO 2 ,
(3) —O—phenyl, which is unsubstituted or substituted with halogen, hydroxyl, C 1-6 alkyl or —O—C 1-6 alkyl,
(4) C 1-6 alkyl, which is unsubstituted or substituted with halogen, hydroxyl or phenyl,
(5) C 3-6 cycloalkyl, which is unsubstituted or substituted with halogen, hydroxyl or phenyl, and
(6) C 2-4 alkenyl, which is unsubstituted or substituted with C 3-6 cycloalkyl or phenyl;
R 5a is independently selected from the group consisting of:
(1) hydrogen,
(2) fluoro,
(3) chloro,
(4) bromo,
(5) hydroxyl,
(6) —CH 3 ,
(7) —CH 2 OH,
(8) —CH 2 CH 3 ,
(9) —CH 2 ═CH 2 ,
(10) —CH 2 CH 2 CH 3 ,
(11) -cyclopropyl,
(12) —OCH 3 ,
(13) —OCH 2 F,
(14) —OCH 2 -cyclopropyl,
(15) —OCH 2 -phenyl,
(16) —OCH 2 CH 3 ,
(17) —OCH 2 CF 3 ,
(18) —OCH 2 CH 2 CH 3 ,
(19) —OCH 2 (C═O)OCH 2 CH 3 ,
(20) —OCH 2 (C═O)NHCH 2 CH 3 ,
(21) —OSO 2 CH 3 , and
(22) —O(C═O)OCH 3 ;
or a pharmaceutically acceptable salt thereof.
2. The method of claim 1 , wherein the compound is a compound of the formula:
or a pharmaceutically acceptable salt thereof.
3. The method of claim 1 , wherein the compound R 1a is phenyl which is unsubstituted or substituted with one or more halogen.
4. The method of claim 1 , wherein the compound R 1a is C 1-6 alkyl.
5. The method of claim 1 , wherein the compound R 1a is isopropyl or tert-butyl.
6. The method of claim 1 , wherein the compound R 1a is isopropyl.
7. The method of claim 1 , wherein the compound R 1a is cyclopropyl.
8. The method of claim 1 , wherein the compound R 5a is —OCH 2 CF 3 .
9. A method for the treatment of schizophrenia which comprises administering to a human patient in need of such treatment an effective amount of a compound which is selected from the group consisting of:
N-[(1R)-1-(5-bromopyridin-2-yl)ethyl]-2-(4-isopropylphenyl)acetamide;
2-(1,1′-biphenyl-4-yl)-N-[(1R)-1-(5-methoxypyridin-2-yl)ethyl]acetamide;

A method for the treatment of schizophrenia which comprises administering to a human patient in need of such treatment an effective amount of a compound which is 2-(4-isopropylphenyl)-N-[(1R)-1-(5-(2,2,2-trifluoroethoxy)pyridin-2-yl)ethyl]acetamide, or a pharmaceutically acceptable salt thereof.
11. The method of claim 10 wherein the compound is 2-(4-isopropylphenyl)-N-[(1R)-1-(5-(2,2,2-trifluoroethoxy)pyridin-2-yl)ethyl]acetamide.
