Abstract

Newly identified mammalian taste-cell-specific G protein-coupled receptors, and the genes and cDNA encoding said receptors are described. Specifically, T1R G protein-coupled receptors active in taste signaling, and the genes and cDNA encoding the same, are described, along with methods for isolating such genes and for isolating and expressing such receptors. Methods for representing taste perception of a particular tastant in a mammal are also described, as are methods for generating novel molecules or combinations of molecules that elicit a predetermined taste perception in a mammal, and methods for simulating one or more tastes. Further, methods for stimulating or blocking taste perception in a mammal are also disclosed.

Claims (13)

1. An isolated nucleic acid selected from the group consisting of: (i) a nucleic acid that is at least 90% identical to a fragment of SEQ ID NO:20 that is at least 100 nucleotides or the complement thereof; (ii) a nucleic acid that encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NO 4, wherein said fragment comprises at least 50 amino acids.

2. The nucleic acid of claim 1 which is at least 90% identical to a fragment of SEQ ID NO:20 that is at least 150 nucleotides or the complement thereof.

3. The nucleic acid of claim 1 which is at least 90% identical to a fragment of SEQ ID NO:20 that is at least 200 nucleotides or the complement thereof.

4. The nucleic acid of claim 1 which is at least 90% identical to a fragment of SEQ ID NO:20 that is at least 250 nucleotides or the complement thereof.

5. The nucleic acid of claim 1 which encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NOs 4, wherein said fragment comprises at least 50 amino acids.

6. The nucleic acid of claim 1 which encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NOs 4, wherein said fragment comprises at least 100 amino acids.

7. The nucleic acid of claim 1 which encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NOs 4, wherein said fragment comprises at least 150 amino acids.

8. The nucleic acid of claim 1 which encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NOs 4, wherein said fragment comprises at least 200 amino acids.

9. The nucleic acid of claim 1 which encodes a polypeptide that is at least 90% identical to a fragment of the T1R polypeptide in SEQ ID NOs 4, wherein said fragment comprises at least 250 amino acids.

10. A vector containing a nucleic acid according to any one of claims 1 - 9 .

11. A cell containing a vector according to claim 10 .

12. The nucleic acid of claim 1 that further comprises a nucleic acid encoding a fragment of a different GPCR comprising the extracellular or transmembrane region thereof.

13. The cell of claim 11 that further expresses a G protein.
