Abstract

[Problems] To provide a novel mixture for assaying a target nucleic acid, characterized by enabling a nucleic acid assay while: 1) requiring no step of diluting the target nucleic acid; 2) requiring no procedure of changing a probe concentration depending on a concentration of the target nucleic acid. [Means for Solving Problems] 1) A mixture which comprises one internal standard nucleic acid and two nucleic acid probes labeled with a fluorescent dye; 2) a mixture for measuring Km value which comprises one internal standard nucleic acid having a partial mutation and one nucleic acid probe labeled with a fluorescent dye; 3) a mixture which comprises one internal standard nucleic acid and one double nucleic acid probe labeled with two fluorescent dyes; and a method for assaying a nucleic acid by making use thereof.

12 . A nucleic acid probe suitable for assaying or detecting a target nucleic acid, said probe comprising at least one fluorescent dye label, wherein said fluorescent dye is selected from the group consisting of 2-oxo-6,8-difluoro-7-hydroxy-2H-1-benzopyran-3-carboxylic acid, TET, TBSF, HEX and rhodamine 6G.

13 . The nucleic acid probe according to claim 12 , wherein said probe has a cytosine as a 3′-end base and is labeled at the 3′-end base with said fluorescent dye.

14 . The nucleic acid probe according to claim 13 , wherein said probe has a base sequence designed such that said probe is hybridizable with said target nucleic acid and said 3′-end cytosine base corresponds to a guanine base in said target nucleic acid.

15 . The nucleic acid probe according to claim 12 , wherein said fluorescent dye is 2-oxo-6,8-difluoro-7-hydroxy-2H-1-benzopyran-3-carboxylic acid.

16 . The nucleic acid probe according to claim 12 , wherein said fluorescent dye is TET.

17 . The nucleic acid probe according to claim 12 , wherein said fluorescent dye is TBSF.

18 . The nucleic acid probe according to claim 12 , wherein said fluorescent dye is HEX.

19 . The nucleic acid probe according to claim 12 , wherein said fluorescent dye is rhodamine 6G.
