Abstract

The invention provides methods and compositions for simultaneously detecting the activation state of a plurality of proteins in single cells using flow cytometry. The invention further provides methods and compositions of screening for bioactive agents capable of coordinately modulating the activity of a plurality of proteins in single cells. The methods and compositions can be used to determine the protein activation profile of a cell for predicting or diagnosing a disease state, and for monitoring treatment of a disease state.

Claims (19)

1. A system for determining activation states of activatable elements in single cells comprising:

configured to detect for detecting, in a single cell, a first labeled activation state-specific binding element that is specific for and binds to an activation state of a first activatable element, and a second labeled activation state-specific binding element that is different from the first labeled activation state-specific binding element and is specific for and binds to an activation state of a second activatable element, wherein the first activatable element and second activatable element each comprise a first activation state and a second activation state, wherein the first activatable element and the second activatable element are different, and wherein the first labeled activation state-specific binding element and the second labeled activation state-specific binding element are differentially labeled and distinguishably detectable;

a computer system comprising a module configured to determine the activation state of first and second activatable elements in the single cell based on the binding to and detection of the differentially labeled and first and second activation state-specific binding elements to the first and second activatable elements in the cell; and

software configured to determine the state of the single cell based on the determined activation states of the first and second activatable elements in the cell.

2. The system according to claim 1 , wherein the first labeled activation-state specific binding element is a first antibody and the second labeled activation-state specific binding element is a second antibody.

3. The system according to claim 2 , wherein the first antibody is specific for a phosphorylation state of the first activatable element, and the second antibody is specific for a phosphorylation state of the second activatable element.

The system according to claim 2 , wherein the first activatable element and second activatable element are each selected from the group consisting of AKT, PI3K, MAPK, p44/42 MAP kinase, TYK2, p38 MAP kinase, PKC, PKA, SAPK, ELK, JNK, cJun, RAS, Raf, MEK 1/2, MEK 3/6, MEK 4/7, ZAP-70, LAT, SRC, LCK, ERK 1/2, Rsk 1, PYK2, SYK, PDK1, GSK3, FKHR, AFX, PLCγ, NF-kB, FAK, CREB, αIIIβ3, FcγRI, BAD, p70S6K, STAT1, STAT2, STAT3, STAT5 and STAT6.

5. The system according to claim 1 , further comprising the first activation-state specific binding element and the second activation-state specific binding element.

6. The system according to claim 5 , wherein the first activation-state specific binding element is a first antibody and the second activation-state specific binding element is a second antibody.

The system according to claim 6 , wherein the first activatable element and second activatable element are selected from the group consisting of AKT, PI3K, MAPK, p44/42 MAP kinase, TYK2, p38 MAP kinase, PKC, PKA, SAPK, ELK, JNK, cJun, RAS, Raf, MEK 1/2, MEK 3/6, MEK 4/7, ZAP-70, LAT, SRC, LCK, ERK 1/2, Rsk 1, PYK2, SYK, PDK1, GSK3, FKHR, AFX, PLCγ, NF-kB, FAK, CREB, αIIIβ3, FcγRI, BAD, p70S6K, STAT1, STAT2, STAT3, STAT5 and STAT6.

8. The system according to claim 5 , wherein the first activation-state specific binding element and the second activation-state specific binding element are labeled, wherein the first label and the second label are distinguishably different.

9. The system according to claim 1 , further comprising a first label for labeling the first activation-state specific binding element and a second label for labeling the second activation-state specific binding element, wherein the first label and the second label are distinguishably different.

10. The system according to claim 1 , wherein the detector comprises a mass spectrometer.

11. The system according to claim 1 , wherein the detector comprises a flow cytometer.

12. The system according to claim 1 , wherein the state of the cell is the activation state of the cell.

13. The system according to claim 1 , wherein the state of the cell is the apoptotic state of the cell.

14. The system according to claim 1 , wherein the state of the cell is the cytokine responsiveness of the cell.

15. The system according to claim 1 , wherein the state of the cell is the cell cycle state of the cell.

16. The system according to claim 1 , further comprising a sample chamber comprising a composition comprising one or more cells, the one or more cells comprising the first labeled activation state-specific binding element and the second activation state-specific binding element.
