Abstract

The present invention relates to small molecule modulators of mTORC1 and mTORC2, syntheses thereof, and intermediates thereto. Such small molecule modulators are useful in the treatment of proliferative diseases (e.g., benign neoplasms, cancers, inflammatory diseases, autoimmune diseases, diabetic retinopathy) and metabolic diseases. Novel small molecules are provided that inhibit one or more of mTORC1, mTORC2, and PI3K-related proteins. Novel methods of providing soluble mTORC1 and mTORC2 complexes are discussed, as well as methods of using the soluble complexes in a high-throughput manner to screen for inhibitory compounds.

Claims (60)

1. A compound of formula I:

wherein:

R 1 is an optionally substituted group selected from the group consisting of 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

each occurrence of R 2 is independently halogen, —NR 2 —OR, —SR, or an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

j is an integer from 1 to 4, inclusive;

R 3 and R 4 are independently hydrogen, hydroxyl, alkoxy, halogen, or optionally substituted C 1-6 aliphatic, with the proviso that R 3 and R 4 are not taken together to form a ring; and

each R is independently hydrogen, an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and C 1-12 heteroaliphatic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; or:

two R on the same nitrogen atom are taken with the nitrogen to form a 4-7-membered heterocyclic ring having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

2. The compound according to claim 1 , wherein R 1 is optionally substituted 6-membered aryl or optionally substituted 6-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

3. The compound according to claim 2 , wherein R 1 is optionally substituted phenyl or pyridyl.

4. The compound according to claim 1 , wherein R 2 is —NR 2 , optionally substituted 6-10-membered aryl, or optionally substituted 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from nitrogen, oxygen, and sulfur.

5. The compound according to claim 4 , wherein R 2 is optionally substituted 6-10-membered aryl.

6. The compound according to claim 4 , wherein R 2 is optionally substituted 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

7. The compound according to claim 1 , wherein j is 1.

8. The compound according to claim 1 , wherein R 3 and R 4 are both hydrogen.

9. The compound according to claim 1 , wherein the compound is of formula II-a:

wherein:

each occurrence of R x is independently halogen, —NR 2 —OR, —SR, —SO 2 NR 2 , or an optionally substituted group selected from the group consisting of 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

m is an integer from 1 to 4; and when m is 2, 3, or 4, two R x substituents may join together to form a 5-7-membered cyclic group or a 5-7-membered heterocyclic group having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and wherein the 5-7-membered cyclic group or 5-7-membered heterocyclic group formed from two R x substituents is optionally substituted by one group selected from the group consisting of 6-10-membered aryl; 5-10-membered heteroaryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; or C 1-12 aliphatic;

each occurrence of R 2 is independently halogen, —NR 2 —OR, —SR, or an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

j is an integer from 1 to 4, inclusive;

R 3 and R 4 are independently hydrogen, halogen, or optionally substituted C 1-6 aliphatic, with the proviso that R 3 and R 4 are not taken together to form a ring; and

each R is independently hydrogen, an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and C 1-12 heteroaliphatic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; or:

two R on the same nitrogen atom are taken with the nitrogen to form a 4-7-membered heterocyclic ring having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

10. The compound according to claim 1 , wherein R 1 is selected from the group consisting of:

each occurrence of R x is independently halogen, —NR 2 —OR, —SR, —SO 2 NR 2 , or an optionally substituted group selected from the group consisting of 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

k is an integer from 1 to 4; and when m is 2, 3, or 4, two R x substituents may join together to form a 5-7-membered cyclic group or a 5-7-membered heterocyclic group having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and wherein the 5-7-membered cyclic group or 5-7-membered heterocyclic group formed from two R x substituents is optionally substituted by one group selected from the group consisting of 6-10-membered aryl; 5-10-membered heteroaryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; or C 1-12 aliphatic;

each occurrence of R 2 is independently halogen, —NR 2 —OR, —SR, or an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

j is an integer from 1 to 4, inclusive;

R 3 and R 4 are independently hydrogen, halogen, or optionally substituted C 1-6 aliphatic, with the proviso that R 3 and R 4 are not taken together to form a ring; and

each R is independently hydrogen, an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and C 1-12 heteroaliphatic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; or:

two R on the same nitrogen atom are taken with the nitrogen to form a 4-7-membered heterocyclic ring having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

12. The compound according to claim 9 , wherein at least one R x is:

R 1 is an optionally substituted group selected from the group consisting of 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur;

R 2 is independently halogen, —NR 2 —OR, —SR, or an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 heteroaliphatic; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and

each R is independently hydrogen, an optionally substituted group selected from the group consisting of C 1-12 acyl; 6-10-membered aryl; C 7-15 arylalkyl; C 6-15 heteroarylalkyl; C 1-12 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and C 1-12 heteroaliphatic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; or:

two R on the same nitrogen atom are taken with the nitrogen to form a 4-7-membered heterocyclic ring having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

15. The compound according to claim 14 , wherein R 2 is 6-10-membered aryl.

16. The compound according to claim 14 , wherein R 2 is 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

17. The compound according to claim 14 , wherein R 2 is selected from:

18. A pharmaceutical composition comprising a compound according to claim 1 and a pharmaceutically acceptable excipient.

19. The compound according to claim 1 , wherein R 1 is optionally substituted 6-10-membered aryl or optionally substituted 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

20. The compound according to claim 3 , wherein R 1 is optionally substituted phenyl.

21. The compound according to claim 20 , wherein R 1 is 3-trifluoromethylphenyl.

22. The compound according to claim 1 , wherein R 2 is —NHR, wherein R is optionally substituted 6-10-membered aryl or optionally substituted 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

23. The compound according to claim 1 , wherein j is 1 or 2.

24. The compound according to claim 1 , wherein one of R 3 and R 4 is hydrogen.

25. The compound according to claim 1 , wherein R 1 is selected from the group consisting of:

The compound according to claim 9 or 11 , wherein each occurrence of R x is independently —NR 2 , —OR, —SO 2 NR 2 , or an optionally substituted group selected from the group consisting of 6-10-membered aryl; C 1-6 aliphatic; 5-10-membered heteroaryl having 1-4 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur; and 4-7-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

27. The compound according to claim 26 , wherein at least one occurrence of R x is NR 2 , wherein two R on the same nitrogen atom are taken with the nitrogen to form a 5-6-membered heterocyclic ring having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

28. The compound according to claim 26 , wherein at least one occurrence of R x is 4-7- membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

29. The compound according to claim 26 , wherein each occurrence of R x is independently —NH 2 , —OR, —SO 2 NR 2 , or an optionally substituted group selected from the group consisting of 6-membered aryl; C 1-4 aliphatic; and 5-6-membered heterocyclic having 1-2 heteroatoms independently selected from the group consisting of nitrogen, oxygen, and sulfur.

30. The compound according to claim 11 , wherein k is 1 or 2.

31. The compound according to claim 26 , wherein k is 2.
