Abstract

The invention includes method of determining if a subject has a genetic predisposition to clinically diagnosed schizophrenia (SZ), schizotypal personality disorder (SPD), and/or schizoaffective disorder (SD).

Claims (20)

1. A method of determining a human subject's risk of developing schizophrenia, the method comprising determining a ceramide kinase gene (CERK) haplotype of the subject, wherein the CERK haplotype comprises alleles of single nucleotide polymorphisms rs135667 and rs1548977, and the subject has an increased risk of developing schizophrenia when the haplotype comprises a “G” allele at said single nucleotide polymorphism rs135667 and an “A” allele at said single nucleotide polymorphism rs1548977, and the subject has a decreased risk of developing schizophrenia when the haplotype comprises a “G” allele at said single nucleotide polymorphism rs135667 and a “G” allele at said single nucleotide polymorphism rs1548977.

2. The method of claim 1 , wherein said determining the CERK haplotype comprises:

obtaining a sample comprising DNA from the subject; and

determining the identities of the alleles of single nucleotide polymorphisms rs135667 and rs1548977.

3. The method of claim 2 , wherein the sample is obtained from the subject by a health care provider.

4. The method of claim 2 , wherein the sample is provided by the subject without the assistance of a health care provider.

5. The method of claim 1 , further comprising determining the presence or absence of one or more additional markers associated with schizophrenia.

6. The method of claim 1 , wherein the subject has one or more risk factors associated with schizophrenia.

7. The method of claim 6 , wherein the one or more of the risk factors associated with schizophrenia include: the subject has a relative with schizophrenia; the subject has eye tracking dysfunction; the subject has deficits in working memory; or the subject has mixed-handedness.

8. The method of claim 6 , wherein the risk factors associated with schizophrenia include: the subject has one or more relatives who have or had schizophrenia and said one or more relatives include grandparents, parents, uncles, aunts, siblings, or children of the subject.

9. The method of claim 1 , wherein the subject is a child or a fetus or an embryo, and one of the relatives of the subject has schizophrenia.

10. A method of determining a human subject's risk of developing schizophrenia, the method comprising determining a ceramide kinase gene (CERK) haplotype of the subject, wherein the CERK haplotype comprises an allele of single nucleotide polymorphism rs1548977, and the subject has an increased risk of developing schizophrenia when the haplotype comprises an “A” allele at said single nucleotide polymorphism rs1548977.

11. The method of claim 10 , wherein said determining the CERK haplotype comprises: obtaining a sample comprising DNA from the subject; and determining the identity of the allele of single nucleotide polymorphism rs1548977.

12. The method of claim 11 , wherein the sample is obtained from the subject by a health care provider.

13. The method of claim 11 , wherein the sample is provided by the subject without the assistance of a health care provider.

14. The method of claim 10 , further comprising determining the presence or absence of one or more additional markers associated with schizophrenia.

15. The method of claim 10 , wherein the subject has one or more risk factors associated with schizophrenia.

16. The method of claim 15 , wherein the one or more of the risk factors associated with schizophrenia include: the subject has a relative with schizophrenia; the subject has eye tracking dysfunction; the subject has deficits in working memory; or the subject has mixed-handedness.

17. The method of claim 15 , wherein the risk factors associated with schizophrenia include: the subject has one or more relatives who have or had schizophrenia and said one or more relatives include grandparents, parents, uncles, aunts, siblings, or children of the subject.

18. The method of claim 10 , wherein the subject is a child or a fetus or an embryo, and one of the relatives of the subject has schizophrenia.
