Abstract

An integrated sample analysis system is disclosed. The sample analysis system contains (1) a sample preparation/analysis module having sample purification device comprising a monolith that binds specifically to nucleic acids and a sample analysis device comprising a microarray enclosed in a reaction chamber having a hydrophilic interior surface; (2) a temperature control module comprising a thermocycler having a thermally conductive temperature-control bladder; and (3) an imaging device capable of capturing an image of the microarray in the reaction chamber.

Claims (27)

1. A disposable reaction cassette, comprising:

a housing comprising a top end and a bottom end, the housing holding at least one reagent container that is pre-packaged with a reagent needed for a sample analysis procedure and is sealed with a pierceable or removable cover at a top end of said reagent container;

a fluidically sealed flow strip comprising:

a pierceable check valve; and

a flow cell comprising one or more polymerase chain reaction chambers, wherein the one or more polymerase chain reaction chambers are fluidically connected to the pierceable check valve,

wherein the top end of the housing is spaced above and apart from the flow strip,

wherein the reagent container is completely segregated from the flow strip,

wherein each polymerase chain reaction chamber consists of a single reservoir that includes a polynucleotide gel element microarray, or a peptide gel element microarray, wherein the gel element is a hemispherical porous hydrophilic polymer,

wherein each polymerase chain reaction chamber has a bottom wall with a bottom interior surface on which the microarray is formed and a top wall with a top interior surface which faces said bottom interior surface, wherein the entire top interior surface is a hydrophilic interior surface that forms a contact angle of 45° or smaller with a drop of pure water resting on said interior surface to facilitate uniform filling of the polymerase chain reaction chamber, and an exterior surface that is configured to allow efficient thermal transfer to the polymerase chain reaction chamber to enable a polymerase chain reaction, wherein the top wall is made from a transparent material that is configured to allow optical interrogation of the microarray in the polymerase chain reaction chamber.

2. The disposable reaction cassette of claim 1 , wherein said flow strip further comprises one or more pin valves that control fluid flow within said flow strip.

3. The disposable reaction cassette of claim 1 , wherein said flow strip further comprises an absorbent.

4. The disposable reaction cassette of claim 3 , wherein said flow strip comprises one or more pin valves, and further wherein said absorbent is in fluid communication with said one or more reaction chambers via through the one or more pin valves.

5. The disposable reaction cassette of claim 1 , further comprising one or more containers pre-packaged with a plurality of lysis beads and a magnetic stirrer.

6. The disposable reaction cassette of claim 1 , wherein said pierceable cover at a top end of said container is a dome valve for loading a liquid sample into said reaction chamber.

7. The disposable reaction cassette of claim 1 , wherein said cassette further comprises a disposable sample purification device.

8. The disposable reaction cassette of claim 7 , wherein said disposable sample purification device is a pipette tip comprising a monolith filter that binds specifically to nucleic acids.

9. The disposable reaction cassette of claim 1 , wherein said reagent container is pre-packaged with a reagent needed for a sample analysis procedure and is sealed with a pierceable cover at a top end of said reagent container.

10. A disposable reaction cassette, comprising:

a housing comprising a top end and a bottom end, the housing holding at least one reagent container that is pre-packaged with a reagent needed for a sample analysis procedure and is sealed with a pierceable or removable cover at a top end of said reagent container;

a fluidically sealed flow strip comprising:

a pierceable check valve; and

a flow cell comprising one or more polymerase chain reaction chambers, wherein the one or more polymerase chain reaction chambers are fluidically connected to the pierceable check valve,

wherein the top end of the housing is spaced above and apart from the flow strip,

wherein the reagent container is completely segregated from the flow strip,

wherein each polymerase chain reaction chamber consists of a single reservoir that includes a polynucleotide gel element microarray, or a peptide gel element microarray, wherein the gel element is a hemispherical porous hydrophilic polymer,

wherein each polymerase chain reaction chamber has a bottom wall with a bottom interior surface on which the microarray is formed and a top wall with a top interior surface which faces said bottom interior surface, wherein the entire top interior surface is a hydrophilic interior surface that forms a contact angle of 45° or smaller with a drop of pure water resting on said interior surface to facilitate uniform filling of the polymerase chain reaction chamber, and an exterior surface that is configured to allow efficient thermal transfer to the polymerase chain reaction chamber to enable a polymerase chain reaction, wherein the top wall is made from a transparent material that is configured to allow optical interrogation of the microarray in the polymerase chain reaction chamber, and

wherein said flow strip further comprises one or more pin valves that control fluid flow within said flow strip, and an absorbent.
