Methods of determining cancer cell responsiveness to a notch inhibitory agent
The present invention is based on the discovery that the Notch signaling pathway is associated with cancer. Accordingly, the invention provides methods and compositions for treating cancer. Also provided are methods of modulating the expression and/or activity of proteins in the Notch signaling pathway for use in diagnoses and treatment of cancer in a subject.
1. A method of determining whether a cancer cell is responsive to treatment by a Notch pathway inhibitory agent wherein the inhibitory agent is a fusion protein comprising a negative regulator of Notch signaling and an Antennapedia homeodomain, the method comprising, (i) administering said Notch inhibitory agent to a cancer cell that has an increased level of Notch 1 as compared to a normal cell and (ii) measuring the level of Notch 1, wherein a lower level of Notch 1 in the cancer cell treated with the Notch inhibitory agent as compared with the level of Notch 1 in a cancer cell not treated with the Notch inhibitory agent is indicative of a cancer cell responsive to treatment by a Notch pathway inhibitory agent.
2. The method of claim 1 , wherein the negative regulator of Notch signaling in the Notch pathway inhibitory agent comprises a dominant negative Mastermind isoform or a Numb isoform.
3. The method of claim 1 , further comprising determining the level of Numb expression in the cancer cell, wherein a low level of Numb expression as compared with the level in a normal cell, is indicative of a cancer cell responsive to treatment by a Notch pathway inhibitory agent.
4. The method of claim 1 , wherein the cancer cell is a breast cancer cell, an ovarian cancer cell, a colon cancer cell, a lung cancer cell, a prostate cancer cell, a hematopoietic cancer cell or a pancreatic cancer cell.
5. The method of claim 1 , wherein the Notch pathway inhibitory agent is set forth in the amino acid sequence of SEQ ID NO: 1 or SEQ ID NO: 2.
