Abstract

The present invention involves the use of transcription factors including Tbx5, Mef2C, Hand2, myocardin and Gata4 to reprogram cardiac fibroblasts into cardiomyocytes, both in vitro and in vivo. Such methods find particular use in the treatment of patients post-myocardial infarction to prevent or limit scarring and to promote myocardial repair.

Claims (21)

1. A method of treating a subject having suffered a myocardial infarct (MI) comprising delivering to the heart of said subject Tbx5, Gata4, Hand2 and Mef2C expression cassettes.

2. The method of claim 1 , further comprising delivering to said subject a myocardin expression cassette.

3. The method of claim 1 , wherein said expression cassettes are comprised in replicable vectors.

4. The method of claim 3 , wherein said replicable vectors are viral vectors.

5. The method of claim 4 , wherein said viral vectors are adenoviral vectors or retroviral vectors.

6. The method of claim 3 , wherein said replicable vectors are non-viral vectors.

7. The method of claim 6 , wherein said non-viral vectors are disposed in a lipid delivery vehicle.

8. The method of claim 1 , wherein expression cassettes are delivered 24 hours to one month following said MI.

9. The method of claim 2 , wherein said myocardin expression cassette is delivered within 24 hours.

10. The method of claim 1 , wherein said expression cassettes expression cassettes are delivered multiple times.

11. The method of claim 10 , wherein said myocardin expression cassette is delivered multiple times.

12. The method of claim 1 , wherein said expression cassettes are delivered 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 26, 27, 28, 29, 30 or 31 times.

13. The method of claim 2 , wherein said myocardin expression cassette is delivered 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, 21, 22, 23, 24, 25, 26, 27, 28, 29, 30 or 31 times.

14. The method of claim 1 , wherein each of said expression cassettes are delivered daily.

15. The method of claim 2 , wherein said myocardin expression cassette is delivered daily.

16. The method of claim 1 , wherein each of said expression cassettes are delivered via intracardiac injection.

17. The method of claim 1 , wherein said subject is further administered oxygen, aspirin, and or nitroglycerin.

18. The method of claim 1 , wherein said subject is further administered percutaneous coronary intervention.

19. The method of claim 1 , wherein said subject is further administered a fibrinolytic.

20. The method of claim 1 , wherein said MI is non-ST-elevated MI.

21. The method of claim 1 , wherein said MI is ST-elevated MI.
