Abstract

Disclosed herein are cohesive soft tissue fillers, for example, dermal and subdermal fillers, based on hyaluronic acids and pharmaceutically acceptable salts thereof. In one aspect, hyaluronic acid-based compositions described herein include a therapeutically effective amount of at least one anesthetic agent, for example, lidocaine. The present hyaluronic acid-based compositions including lidocaine have an enhanced stability and cohesivity, relative to conventional compositions including lidocaine, for example when subjected to sterilization techniques or when stored for long periods of time. Methods and processes of preparing such hyaluronic acid-based compositions are also provided.

Claims (33)

1. A dermal filler composition comprising hyaluronic acid (HA) crosslinked with 1,4-butanediol diglycidyl ether (BDDE), and about 0.3% lidocaine by weight,

wherein the lidocaine is freely released in vivo; and

wherein the composition is sterile.

2. The composition of claim 1 , wherein the composition further comprises free HA.

3. The composition of claim 2 , wherein the total amount of crosslinked HA and free HA is about 20 mg/mL to about 30 mg/mL.

4. The composition of claim 1 wherein the hyaluronic acid crosslinked with BDDE is in the form of particles.

5. The composition of claim 4 wherein the particles have an average size of at least about 200 μm.

6. The composition of claim 4 wherein the particles have an average size of at least about 250 μm.

7. The composition of claim 4 wherein the particles have an average size of less than about 200 μm.

8. The composition of claim 1 wherein the hyaluronic acid crosslinked with BDDE has a degree of crosslinking between about 2% and about 20%.

9. The composition of claim 1 wherein the hyaluronic acid crosslinked with BDDE has a degree of crosslinking of less than about 5%.

10. The composition of claim 1 , wherein the pH is from about 7 to about 7.5.

11. The composition of claim 10 , wherein the pH is about 7.

12. The composition of claim 1 , wherein the composition is sterilized by autoclave.

13. The composition of claim 1 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 3 months.

14. The composition of claim 1 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 6 months.

15. The composition of claim 1 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 9 months.

16. The composition of claim 1 , having a viscosity that is substantially constant during storage under ambient conditions for at least 3 months.

17. The composition of claim 1 , having a viscosity that is substantially constant during storage under ambient conditions for at least 6 months.

18. The composition of claim 1 , having a viscosity that is substantially constant during storage under ambient conditions for at least 9 months.

19. The composition of claim 1 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 3 months.

20. The composition of claim 1 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 6 months.

21. The composition of claim 1 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 9 months.

22. The composition of claim 1 , wherein the composition is sterilized by heat sterilization between about 120° C. and about 130° C.

23. The composition of claim 22 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 3 months.

24. The composition of claim 22 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 6 months.

25. The composition of claim 22 , having an extrusion force that is substantially constant during storage under ambient conditions for at least 9 months.

26. The composition of claim 22 , having a viscosity that is substantially constant during storage under ambient conditions for at least 3 months.

27. The composition of claim 22 , having a viscosity that is substantially constant during storage under ambient conditions for at least 6 months.

28. The composition of claim 22 , having a viscosity that is substantially constant during storage under ambient conditions for at least 9 months.

29. The composition of claim 22 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 3 months.

30. The composition of claim 22 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 6 months.

31. The composition of claim 22 , wherein the lidocaine does not substantially degrade the HA during storage under ambient conditions for at least 9 months.
