Abstract

A multi-dimensional liquid analysis system includes a flow splitter for separating mobile phase outflow from a first dimension liquid analysis system into first and second liquid split outlet flows. Volumetric flow rate control of the split outlet flows is provided by a flow control pump which withdraws one of the split outlet flows from the flow splitter at a controlled withdrawal flow rate to define the other split outlet flow rate as the difference between the outflow rate from the first dimension system and the withdrawal flow rate. In this manner, accurate and consistent flow division can be accomplished, which is particularly useful for multi-dimensional liquid analysis.

Claims (69)

1. A multi-dimensional liquid analysis system, comprising:

a first separation system including a first separation column, the first separation column configured to chromatographically separate a liquid mobile phase into a first dimension outflow having a first outflow rate;

a flow splitter coupled with the first dimension outflow, the flow splitter configured to separate the first dimension outflow into a first split outlet flow and a second split outlet flow;

a second separation system including a second separation column, the second separation column configured to chromatographically separate the second split outlet flow; and

a flow control pump fluidly coupled to the flow splitter and configured to withdraw the second split outlet flow from the flow splitter at a controlled withdrawal flow rate to obtain a representative sampling of compounds in the first dimension outflow for separation of the second split outlet flow in the second separation system.

2. The multi-dimensional liquid analysis system as recited in claim 1 , including a flow restrictor restricting the first split outflow to create a fluid pressure at the flow splitter of 1-1000 kilopascals.

3. The multi-dimensional liquid analysis system as recited in claim 1 , wherein the flow control pump is a positive displacement pump arranged to establish the second split outlet flow while operating in a negative displacement mode.

4. The multi-dimensional liquid analysis system as recited in claim 3 , wherein the flow control pump is a syringe pump.

The multi-dimensional liquid analysis system as recited in claim 1 , further comprising a multiple-port injection valve configured to inject the second split outlet flow into the second separation column, a first sample loop defining a first flow path of the second split outlet flow to the second separation column.

The multi-dimensional liquid analysis system as recited in claim 5 , wherein the injection valve includes a second sample loop having a volume, the second sample loop defining a second flow path of the second split outlet flow to the second separation column that is distinct from the first flow path.

7. A multi-dimensional liquid analysis system, comprising:

a first separation system including a first separation column configured to chromatographically separate a liquid mobile phase into a first dimension outflow having a first outflow rate;

a flow splitter configured to separate the first dimension outflow into a first split outlet flow and a second split outlet flow;

a second separation system including a second separation column configured to chromatographically separate the second split outlet flow;

a flow control pump fluidly coupled to the flow splitter and configured to withdraw the second split outlet flow from the flow splitter at a controlled withdrawal flow rate to obtain a representative sampling of compounds in the first dimension outflow; and

the flow control pump is arranged to establish the second split outlet flow while operating in a negative displacement mode.

8. The multi-dimensional liquid analysis system as recited in claim 7 , including a flow restrictor restricting the first split outflow to create a fluid pressure at the flow splitter of 1-1000 kilopascals.

9. The multi-dimensional liquid analysis system as recited in claim 7 , wherein the flow control pump is a positive displacement pump arranged to establish the second split outlet flow while operating in a negative displacement mode.

10. The multi-dimensional liquid analysis system as recited in claim 9 , wherein the flow control pump is a syringe pump.

11. The multi-dimensional liquid analysis system as recited in claim 7 , further comprising a multiple-port injection valve configured to inject the second split outlet flow into the second separation column, a first sample loop defining a first flow path of the second split outlet flow to the second separation column.

The multi-dimensional liquid analysis system as recited in claim 11 , wherein the injection valve includes a second sample loop having a volume, the second sample loop defining a second flow path of the second split outlet flow to the second separation column that is distinct from the first flow path.

13. A multi-dimensional liquid analysis system, comprising:

a first separation system including a first separation column for chromatographically separating a liquid mobile phase into a first dimension outflow having a first outflow rate;

a flow splitter for separating the first dimension outflow into a first split outlet flow and a second split outlet flow;

a second separation system including a second separation column for chromatographically separating the second split outlet flow; and

a flow control pump fluidly coupled to the flow splitter and configured to withdraw the second split outlet flow from the flow splitter at a controlled withdrawal flow rate to obtain a representative sampling of compounds in the first dimension outflow, wherein the controlled withdrawal flow rate defines a first split outlet flow rate as the difference between the first outflow rate and the withdrawal flow rate, wherein the withdrawal flow rate is controlled by:

wherein,

F c =controlled flow rate at syringe pump flow stream;

V L =volume of sample loop;

T 2a =analysis time of second dimension chromatograph; and

T 2e =equilibration time of second dimension.

14. The multi-dimensional liquid analysis system as recited in claim 13 , including a flow restrictor restricting the first split outflow to create a fluid pressure at the flow splitter of 1-1000 kilopascals.

15. The multi-dimensional liquid analysis system as recited in claim 13 , wherein the controlled withdrawal flow rate is suitable for separation of the second split outlet flow in the second separation system.

16. The multi-dimensional liquid analysis system as recited in claim 15 , wherein the flow control pump is a positive displacement pump arranged to establish the second split outlet flow while operating in a negative displacement mode.

The multi-dimensional liquid analysis system as recited in claim 13 , further comprising a multiple-port injection valve configured to inject the second split outlet flow into the second separation column, the first sample loop defining a first flow path of the second split outlet flow to the second separation column.

The multi-dimensional liquid analysis system as recited in claim 17 , wherein the injection valve includes a second sample loop having a volume, the second sample loop defining a second flow path of the second split outlet flow to the second separation column that is distinct from the first flow path.

19. A multi-dimensional liquid analysis system, comprising:

a first separation system including a first separation column for chromatographically separating a liquid mobile phase into a first dimension outflow having a first outflow rate;

a flow splitter for separating the first dimension outflow into a first split outlet flow and a second split outlet flow;

a second separation system including a second separation column for chromatographically separating the second split outlet flow; and

a flow control pump fluidly coupled to the flow splitter and configured to withdraw the second split outlet flow from the flow splitter at a controlled withdrawal flow rate to obtain a representative sampling of compounds in the first dimension outflow, wherein the controlled withdrawal flow rate defines a first split outlet flow rate as the difference between the first outflow rate and the withdrawal flow rate, wherein the withdrawal flow rate is controlled by:

20. The multi-dimensional liquid analysis system as recited in claim 19 , wherein the controlled withdrawal flow rate is suitable for separation of the second split outlet flow in the second separation system.

21. The multi-dimensional liquid analysis system as recited in claim 19 , wherein the flow control pump is a positive displacement pump arranged to establish the second split outlet flow while operating in a negative displacement mode.

22. The multi-dimensional liquid analysis system as recited in claim 19 , including a flow restrictor restricting the first split outflow to create a fluid pressure at the flow splitter of 1-1000 kilopascals.

23. The multi-dimensional liquid analysis system as recited in claim 19 , further comprising a multiple-port injection valve configured to inject the second split outlet flow into the second separation column, the first sample loop defining a first flow path of the second split outlet flow to the second separation column.

The multi-dimensional liquid analysis system as recited in claim 23 , wherein the injection valve includes a second sample loop having a volume, the second sample loop defining a second flow path of the second split outlet flow to the second separation column that is distinct from the first flow path.
