Abstract

Compositions of and methods for using synthetic retinal derivatives as retinoid replacements and opsin agonists are provided.

Claims (31)

1. A pharmaceutical ophthalmological composition for use in treating a visual disorder due to a deficiency of 11-cis-retinal in an eye of a human subject, comprising:

a synthetic retinyl ester that is a 9-cis-retinyl ester or an 11-cis-retinyl ester, wherein when the synthetic retinyl ester is administered to the human subject a functional opsin/chromophore complex is formed in the eye; and,

a pharmaceutically acceptable vehicle;

wherein the 9-cis-retinyl ester has a structure as represented by Formula I or the 11-cis-retinyl ester has a structure as represented by Formula II, wherein A in each formula is CH 2 OR and R forms a retinyl ester, and the ester substituent of said retinyl ester comprises a carboxylate radical of a C 1 to C 10 monocarboxylic acid or a C 2 to C 22 polycarboxylic acid:

wherein the ester substituent is selected from the group consisting of formate, acetate, propionate, butyrate, valerate, caproate, heptanoate, caprylate, pelargonate, caprate, oxalate, malonate, succinate, fumarate, malate, glutarate, adipate, pimelate, suberate, azelate, sebacate, citrate, oxaloacetate, and ketoglutarate.

The pharmaceutical ophthalmological composition of claim 1 , wherein the endogenous 11-cis-retinal deficiency is associated with Retinitis Pigmentosa (RP), Leber Congenital Amaurosis (LCA), Retinitis Punctata Albesciens, Congenital Stationary Night Blindness, Fundus Albipunctatus, Age-Related Macular Degeneration, loss of night vision or contrast sensitivity.

3. The pharmaceutical ophthalmological composition of claim 1 , wherein the human subject has a mutation selected from an RPE65 mutation and an LRAT mutation.

4. The pharmaceutical ophthalmological composition of claim 1 , wherein the composition is formulated for local administration to an eye of a human subject.

5. The pharmaceutical ophthalmological composition of claim 1 , wherein the composition is formulated for oral administration, systemic administration, time release administration, controlled release administration or prophylactic administration.

6. The pharmaceutical ophthalmological composition of claim 1 , wherein the composition is formulated for oral administration and the pharmaceutically acceptable vehicle is canola oil.

7. The pharmaceutical ophthalmological composition of claim 5 , wherein the oral administration is in a unit dosage of about 1.0 to about 1000 mg.

8. The pharmaceutical ophthalmological composition of claim 4 , wherein the local administration is topically, by eye drops, intraocular injection, or periocular injection.

9. The pharmaceutical ophthalmological composition of claim 5 , wherein the systemic administration is orally, intravenous injection or intramuscular injection.

10. The pharmaceutical ophthalmological composition of claim 1 , wherein the composition is formulated in a form of an implant, a microencapsulation delivery system, a liquid or a biodegradable polymer.

11. The pharmaceutical ophthalmological composition of claim 1 , wherein the synthetic retinyl ester is 9-cis-retinyl succinate or 9-cis-retinyl acetate.

12. A pharmaceutical ophthalmological composition for the use in treating a deficiency of 11-cis-retinal due to an inherited mutation in RPE65 or BRAT in an eye of a human subject, comprising:

a synthetic retinal ester that is a 9-cis-retinyl ester or an 11-cis-retinyl ester, wherein the 9-cis-retinyl ester has a structure as represented by Formula I or the 11-cis-retinyl ester has a structure as represented by Formula II, wherein A in each formula is CH 2 OR and R forms a retinyl ester, and the ester substituent of said retinyl ester comprises a carboxylate radical of a C 1 to C 10 monocarboxylic acid or a C 2 to C 22 polycarboxylic acid:

wherein the ester substituent is selected from the group consisting of formate, acetate, propionate, butyrate, valerate, caproate, heptanoate, caprylate, pelargonate, caprate, oxalate, malonate, succinate, fumarate, malate, glutarate, adipate, pimelate, suberate, azelate, sebacate, citrate, oxaloacetate, and ketoglutarate; and,

a pharmaceutically acceptable vehicle.

The pharmaceutical ophthalmological composition of claim 12 , wherein the endogenous 11-cis-retinal deficiency is associated with Retinitis Pigmentosa (RP), Leber Congenital Amaurosis (LCA), Retinitis Punctata Albesciens, Congenital Stationary Night Blindness, Fundus Albipunctatus, Age-Related Macular Degeneration, loss of night vision or contrast sensitivity.

14. The pharmaceutical ophthalmological composition of claim 1 , wherein the synthetic retinyl ester is 11-cis-retinyl succinate or 11-cis-retinyl acetate.

15. The pharmaceutical ophthalmological composition of claim 1 , wherein the ester substituent is selected from the group consisting of succinate, citrate, ketoglutarate, fumarate, malate, and oxaloacetate.

16. The pharmaceutical ophthalmological composition of claim 5 , wherein the oral administration is in a unit dosage of about 10 to about 250 mg of the retinyl ester.

17. The pharmaceutical ophthalmological composition of claim 5 , wherein the composition is in a form selected from a tablet, a pill, a sachet, or a capsule.

18. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition is formulated as an oral dosage form.

19. The pharmaceutical ophthalmological composition of claim 18 , wherein the oral dosage form is a tablet, a pill, a sachet or a capsule.

20. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition comprises an oral dose of about 1.0 to about 1000 mg of the synthetic retinyl ester.

21. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition comprises an oral dose of about 10 to about 250 mg of the synthetic retinyl ester.

22. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition is formulated for injection.

23. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition is formulated for intravenous or intramuscular injection.

24. The pharmaceutical ophthalmological composition of claim 12 , wherein the composition is formulated as a time release formulation, as a controlled release formulation, as an implant, as a microencapsulated delivery system, or with a biodegradable and biocompatible polymer.
