Abstract

The invention provides a screening method for identifying an insect-specific TRPA1 modulator by comparing modulation of an insect TRPA1 and a mammalian TRPA1. The invention further provides method of insect control by applying to an insect a insect-specific TRPA1 modulator identified by the screening method.

Claims (10)

1. A method of identifying an insect-specific transient receptor potential ion channel A1 (TRPA1) modulator comprising:

(a) contacting a test compound with an insect TRPA1 and a vertebrate TRPA1, wherein the test compound is a reactive electrophile; and

(b) assaying modulation of insect and vertebrate TRPA1 activity, wherein if modulation of the activity of insect TRPA1 is increased by at least 10% relative to modulation of activity of the vertebrate TRPA, the test compound is identified as an insect-specific TRPA1 modulator.

2. The method of claim 1 , wherein the compound inhibits the activity of the insect TRPA1.

3. The method of claim 1 , wherein the compound activates the insect TRPA1.

The method of claim 1 , wherein the insect TRPA1 is selected from the group consisting of D. melanogaster TRPA1 isoform F (Accession No. ABW08500.3) (SEQ ID NO: 1), D. melanogaster TRPA1 isoform E (Accession No. AAF50356.4) (SEQ ID NO: 2), D. melanogaster TRPA1 isoform F (Accession No. NP_001097554.3) (SEQ ID NO: 3), D. melanogaster TRPA1 isoform E (Accession No. NP_648263.4) (SEQ ID NO: 4), D. melanogaster TRPA1 (Accession No. Q7Z020.3) (SEQ ID NO: 5), Anopheles gambiae TRPA1 (Accession No. ACC86138.1) (SEQ ID NO: 6), and Tribolium castaneum hypothetical protein TcasGA2_TC002449 (Accession No. EFA01253.1) (SEQ ID NO: 7).

The method of claim 1 , wherein the vertebrate TRPA1 is selected from the group consisting of Rattus norvegicus transient receptor potential cation channel, subfamily A, member 1 (Accession No. NP_997491.1) (SEQ ID NO: 8); Rattus norvegicus transient receptor potential cation channel subfamily A member 1 (Accession No. AAS78661.1) (SEQ ID NO: 9); Mus musculus transient receptor potential cation channel, subfamily A, member 1 (Accession No. NP_808449.1) (SEQ ID NO: 10); Homo sapiens transient receptor potential cation channel subfamily A member 1 (Accession No. NP_015628.2) (SEQ ID NO: 11); Mus musculus transient receptor potential cation channel, subfamily A, member 1 (Accession No. AAI31964.1) (SEQ ID NO: 12); Mus musculus transient receptor potential cation channel, subfamily A, member 1 (Accession No. AAI20564.1) (SEQ ID NO: 13); Mus musculus transient receptor potential cation channel, subfamily A, member 1, isoform CRA_b (Accession No. EDL14332.1) (SEQ ID NO: 14); Mus musculus transient receptor potential cation channel, subfamily A, member 1, isoform CRA_a (Accession No. EDL14331.1) (SEQ ID NO: 15); Bos Taurus transformation sensitive protein p120 (Accession No. XP_581588.2) (SEQ ID NO: 16); Pan troglodytes predicted ankyrin-like protein 1 (Accession No. XP_519806.2) (SEQ ID NO: 17); Macaca mulatta predicted ankyrin-like protein 1 (Accession No. XP_001083172.1) (SEQ ID NO: 18); Gallus gallus predicted similar to transient receptor potential cation channel subfamily A member 1 (Accession No. XP_418294.2) (SEQ ID NO: 19); Danio rerio TRPA1 (Accession No. AAV37177.1) (SEQ ID NO: 20); and Danio rerio transient receptor potential cation channel, subfamily A, member 1a (Accession No. NP_001007066.1) (SEQ ID NO: 21).

6. The method claim 1 , wherein the insect TRPA1 or the vertebrate TRPA1 is inside a cell.

7. The method of claim 6 , wherein the cell is an oocyte.

8. The method of claim 1 , wherein the test compound is extracted from a bacteria, plant, fungi, animal cell, or animal tissue.
