Abstract

This document provides methods and materials related to genetic variations of neurological disorders. For example, this document provides methods for using such genetic variations to assess susceptibility of developing Parkinson's disease.

Claims (22)

1. A method of hybridizing a nucleic acid probe comprising:

(a) hybridizing a nucleic acid probe with a detectable label to a polynucleic acid by nucleic acid hybridization or microarray analysis; wherein the polynucleic acid is from a sample from a human subject that has Parkinson's Disease; and

(b) detecting a genetic variation in the polynucleic acid by nucleic acid hybridization or microarray analysis by detecting the detectable label of the nucleic acid probe, wherein the genetic variation is a copy number variation (CNV) of SEQ ID NO: 1 or the complement thereof; and wherein the CNV is the gain of SEQ ID NO:1 or the complement thereof in ATRNL1 (Attractin-Like 1) gene.

2. The method of claim 1 , wherein the CNV is the gain of SEQ ID NO: 1.

3. The method of claim 1 , wherein the detectable label is a fluorescent label.

4. The method of claim 1 , wherein the method comprises purifying the polynucleic acid.

5. The method of claim 1 , wherein the polynucleic acid is isolated.

6. The method of claim 1 , wherein the polynucleic acid comprises cDNA.

7. The method of claim 1 , wherein the polynucleic acid is from a sample of blood, saliva, urine, tears, skin, tissue, or hair from the human subject.

8. The method of claim 1 , wherein the microarray analysis is selected from the group consisting of a Comparative Genomic Hybridization (CGH) array analysis and an SNP array analysis.

9. The method of claim 8 , wherein the microarray analysis comprises Comparative Genomic Hybridization (CGH) array analysis.

10. The method of claim 1 , further comprising evaluating the human subject's motor skills, autonomic function, neuropsychiatry, mood, cognition, behavior, thoughts, ability to sense, or family history of Parkinson's Disease.

11. A method of synthesizing a nucleic acid product comprising:

(a) synthesizing a nucleic acid product from a polynucleic acid by PCR or sequencing; wherein the polynucleic acid is from a sample from a human subject that has Parkinson's Disease; and

(b) detecting a genetic variation in the nucleic acid product by PCR or sequencing, wherein the genetic variation is a copy number variation (CNV) of SEQ ID NO: 1 or the complement thereof; and wherein the CNV is the gain of SEQ ID NO:1 or the complement thereof in ATRNL1 gene.

12. The method of claim 11 , wherein the CNV is the gain of SEQ ID NO: 1.

13. The method of claim 11 , wherein the method comprises purifying the polynucleic acid.

14. The method of claim 11 , wherein the nucleic acid product synthesized from the polynucleic acid is cDNA.

15. The method of claim 11 , wherein the whole genome or the whole exome of the human subject is analyzed.

16. The method of claim 11 , wherein the polynucleic acid is from a sample of blood, saliva, urine, tears, skin, tissue, or hair from the human subject.

17. The method of claim 11 , wherein the sequencing is a high-throughput sequencing method.

18. The method of claim 11 , further comprising evaluating the human subject's motor skills, autonomic function, neuropsychiatry, mood, cognition, behavior, thoughts, ability to sense, or family history of Parkinson's Disease.
