Abstract

Disclosed are two PTH analog ligands, SP-PTH-AAK and Aib-SP-PTH-AAK, that have long-acting activity at the PTH receptor, as demonstrated both in vitro and in vivo. These polypeptides are thus particularly useful in the treatment of diseases, such as hypoparathyroidism, in which long-acting activity is desired. The method of making the analog polypeptides is also disclosed.

Claims (55)

1. A polypeptide comprising formula (I):

X 01 -Val-X 03 -Glu-Ile-Gln-Leu-X 08 -His-X 10 -X 11 -X 12 -X 13 -

X 14 -X 15 -X 16 -X 17 -X 18 -Arg-Arg-Arg-X 22 -Phe-Leu-X 25 -X 26 -

Leu-Ile-Ala-Glu-Ile-His-Thr-Ala-Glu-Ile

X 01 is Ser or Ala;

X 03 is Ser, Ala, or Aib;

X 08 is Met, Leu, or Nle;

X 10 is Asn or Gln;

X 11 is Leu, Arg, or Har;

X 12 is Gly or Ala;

X 13 is Lys;

X 14 is His or Trp;

X 15 is Ile;

X 16 is Gln;

X 17 is Asp;

X 18 is Ala;

X 22 is Ala;

X 25 is His; and

X 26 is Lys;

or a pharmaceutically acceptable salt thereof.

2. The polypeptide of claim 1 , wherein X 01 and X 03 are Ala; X 10 is Gln; X 11 is Arg; X 12 is Ala; and X 14 is Trp.

3. The polypeptide of claim 1 , wherein X 01 is Ala; X 03 is Aib; X 10 is Gln; X 11 is Har; X 12 is Ala; and X 14 is Trp.

4. The polypeptide of claim 1 , wherein said polypeptide is fewer than 50 amino acids in length.

5. The polypeptide of claim 1 , or a pharmaceutically acceptable salt thereof, comprising the amino acid sequence:

Lys-Trp-Ile-Gln-Asp-Ala-Arg-Arg-Arg-Ala-Phe-Leu-

6. The polypeptide of claim 1 , or a pharmaceutically acceptable salt thereof, comprising the amino acid sequence:

Lys-Trp-Ile-Gln-Asp-Ala-Arg-Arg-Arg-Ala-Phe-Leu-

7. The polypeptide of claim 1 , or a pharmaceutically acceptable salt thereof, comprising an amino acid sequence selected from the group consisting of:

8. A pharmaceutical composition comprising a polypeptide of claim 1 and a pharmaceutically acceptable carrier.

A method for treating a subject having a disease selected from the group consisting of hypoparathyroidism, hyperphosphatemia, osteoporosis, fracture repair, osteomalacia, arthritis, and thrombocytopenia, said method comprising administering a polypeptide of claim 1 to said subject in an amount sufficient to treat said disease.

10. The method of claim 9 , wherein the route of administration is selected from the group consisting of subcutaneously, intravenously, intranasally, transpulmonarily, transdermally, and orally.
