Abstract

The invention provides compositions of novel high penetration compositions (HPC) or high penetration prodrugs (HPP) of 1H-imidazo[4,5-c]quinolin-4-amines and 1H-imidazo[4,5-c]quinolin-4-amine-related compounds, which are capable of crossing biological barriers with high penetration efficiency. The HPPs are capable of being converted to parent active drugs or drug metabolites after crossing the biological barrier and thus can render treatments for the conditions that the parent drugs or metabolites can. Additionally, the HPPs are capable of reaching areas that parent drugs may not be able to access or to render a sufficient concentration at the target areas and therefore render novel treatments. The HPPs can be administered to a subject through various administration routes, e.g., locally delivered to an action site of a condition with a high concentration or systematically administered to a biological subject and enter the general circulation with a faster rate.

Claims (62)

1. A method for delivering a functional unit across a skin barrier in a biological subject, comprising topically administrating to the biological subject a compound of Structure L-1:

F-L 4 -L 2 -T   Structure L-1

including stereoisomers and pharmaceutically acceptable salts thereof, wherein:

F is selected from Structure F, Structure F-1, Structure F-3, Structure F-4, Structure F-5, Structure F-6, Structure F-7, Structure F-8, Structure F-9, Structure F-10, Structure F-11, Structure F-12, Structure F-13, Structure F-14, Structure F-16, Structure F-17, and Structure F-18:

R 4 is selected from hydrogen, R b , —R b OH, —R a OR b , —R a OC(═O)R b , and —R a C(═O)OR b ;

R 5 is selected from hydrogen, R b , —R b OH, —R a OR b , —R a OC(═O)R b , —R a C(═O)R b and —R a C(═O)OR b ;

R 6 is selected from hydrogen, substituted and unsubstituted alkyl, substituted and unsubstituted alkoxyl, and halogen;

R 7 is selected from hydrogen, substituted and unsubstituted alkyl, substituted and unsubstituted alkoxy and halogen;

R a is selected from nothing, substituted and unsubstituted alkyl, and substituted and unsubstituted aryl group;

R b is selected from substituted and unsubstituted alkyl, and substituted and unsubstituted aryl group; and

T is selected from Structure Na, Structure Nb, Structure Nc, Structure Nd, Structure Ne, Structure Nf, Structure Ng, Structure Nh, Structure Ni, Structure Nj, Structure Nk, Structure Nl, Structure Nm, Structure Nn, Structure No, Structure Np, Structure Nq, and Structure Nr:

each R 11 -R 16 is independently selected from nothing, H, substituted and unsubstituted alkyl, substituted and unsubstituted cycloalkyl, substituted and unsubstituted heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted alkoxyl, substituted and unsubstituted alkylthio, substituted and unsubstituted alkylamino, substituted and unsubstituted perfluoroalkyl, and substituted and unsubstituted alkyl halide;

L 2 is selected from nothing, O, S, —N(L 3 )-, —N(L 3 )-CH 2 —O, —N(L 3 )—CH 2 —N(L 5 )—, —O—CH 2 —O—, —O—CH(L 3 )—O—, —O-L 3 -, —N-L 3 -, —S-L 3 -, —N(L 3 )-L 5 - and L 3 ;

L 4 is selected from C═O, C═S,

for each L 2 and L 4 , L 3 and L 5 are independently selected from nothing, H, CH 2 C(═O)OL 6 , substituted and unsubstituted alkyl, substituted and unsubstituted cycloalkyl, substituted and unsubstituted heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted alkoxyl, substituted and unsubstituted alkylthio, substituted and unsubstituted alkylamino, substituted and unsubstituted perfluoroalkyl, and substituted and unsubstituted alkyl halide, wherein any carbon or hydrogen may be further independently replaced with O, S, or P;

L 6 is selected from H, OH, Cl, F, Br, I, substituted and unsubstituted alkyl, substituted and unsubstituted cycloalkyl, substituted and unsubstituted heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted alkoxyl, substituted and unsubstituted alkylthio, substituted and unsubstituted alkylamino, substituted and unsubstituted perfluoroalkyl, and substituted and unsubstituted alkyl halide, wherein any carbon or hydrogen may be further independently replaced with O, S, N, P(O)OL 7 , CH═CH, C≡C, CHL 7 , aryl, heteroaryl, or cyclic groups; and

L 7 is selected from H, OH, Cl, F, Br, I, substituted and unsubstituted alkyl, substituted and unsubstituted cycloalkyl, substituted and unsubstituted heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted alkoxyl, substituted and unsubstituted alkylthio, substituted and unsubstituted alkylamino, substituted and unsubstituted perfluoroalkyl, and substituted and unsubstituted alkyl halide, wherein any carbon or hydrogen may be further independently replaced with O, S, N, CH═CH, C≡C, aryl, heteroaryl, or cyclic groups.

2. The method according to claim 1 , wherein the compound is administered to the biological subject in a pharmaceutical composition comprising said compound or a stereoisomer or pharmaceutically acceptable salt thereof and a pharmaceutically acceptable carrier.

3. The method according to claim 2 , further comprising treating a condition selected from genital wart, perianal wart, infections caused by herpes simplex virus, skin cancer, superfacial basal cell carcinoma, basal cell carcinoma, and squamous cell skin cancer.

4. The method according to claim 1 , further comprising treating a condition selected from genital wart, perianal wart, infections caused by herpes simplex virus, skin cancer, superfacial basal cell carcinoma, basal cell carcinoma, and squamous cell skin cancer.

5. The method according to claim 1 , wherein

R 4 is selected from hydrogen, R b , —R b OH, —R a OR b , —R a OC(═O)R b , and —R a C(═O)OR b ;

R 5 is selected from hydrogen, R b , —R b OH, —R a OR b , —R a OC(═O)R b , —R a C(═O)R b and —R a C(═O)OR b ;

R 6 is selected hydrogen, C 1-12 alkyl, C 1-12 alkoxyl, and halogen;

R 7 is selected from hydrogen, C 1-12 alkyl, C 1-12 alkoxy, and halogen;

R a is selected from nothing, C 1-12 alkyl, and aryl;

R b is C 1-12 alkyl or aryl;

each R 11 -R 16 is independently selected from nothing, H, C 1-12 alkyl, C 3-12 cycloalkyl, C 2-12 heterocycloalkyl, aryl, heteroaryl, C 1-12 alkoxyl, C 1-12 alkylthio, C 1-12 alkylamino, C 1-12 perfluoroalkyl, and C 1-12 alkyl halide;

for each L 2 and L 4 , L 3 and L 5 are independently selected from nothing, H, CH 2 C(═O)OL 6 , C 1-12 alkyl, C 3-12 cycloalkyl, C 2-12 heterocycloalkyl, aryl, heteroaryl, C 1-12 alkoxyl, C 1-12 alkylthio, C 1-12 alkylamino, C 1-12 perfluoroalkyl, and C 1-12 halide;

L 6 is selected from H, OH, Cl, F, Br, I, C 1-12 alkyl, C 3-12 cycloalkyl, C 2-12 heterocycloalkyl, aryl, heteroaryl, C 1-12 alkoxyl, C 1-12 alkylthio, C 1-12 alkylamino, C 1-12 perfluoroalkyl, and C 1-12 alkyl halide.

6. The method according to claim 1 , wherein

F is selected from Structure F-1, Structure F-3, Structure F-4, Structure F-5, Structure F-6, Structure F-7, Structure F-8, Structure F-9, Structure F-10, Structure F-11, Structure F-12, Structure F-13, Structure F-14, Structure F-16, Structure F-17, and Structure F-18:

T is Structure Na:

wherein R 11 and R 12 are independently selected from H, C 1-12 alkyl, C 1-12 alkoxyl, C 1-12 alkylthio, C 1-12 alkylamino, C 1-12 perfluoroalkyl, and C 1-12 alkyl halide;

or T is Structure Nc or Structure Ng:

wherein each R 11 -R 13 is independently C 1-12 alkyl and R 14 is H or C 1-12 alkyl;

L 2 is selected from nothing, O, S, —O—CH 2 —O—, —O-L 3 -, —N-L 3 -, —S-L 3 -, and L 3 ;

L 4 is C═O or C═S; and

L 3 is selected from C 1-12 alkyl, C 1-12 alkoxyl, C 1-12 alkylthio, C 1-12 alkylamino, C 1-12 perfluoroalkyl, and C 1-12 alkyl halide.

7. The method according to claim 6 , wherein:

T is Structure Na:

R 11 and R 12 are independently selected from H and C 1-12 alkyl;

L 2 is C 1-12 alkyl; and

L 4 is C═O.

8. The method according to claim 1 , wherein the compound of Structure L-1 or a pharmaceutically acceptable salt thereof is

sarcosine 1-isobutyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-butyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-benzyl-1H-imidazo[4,5-c]quinolin-4-amide;

N,N-dimethylglycine 1-butyl-1H-imidazo[4,5-c]quinolin-4-amide;

N,N-dimethylglycine 1-n-hexyl-1H-imidazo[4,5-c]quinolin-4-amide;

N,N-dimethylglycine 1-isobutyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-methyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1,2,8-trimethyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-(2-hydroxyethyl)-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1,8-dimethyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1,2-dimethyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-(2,3-dihydroxypropyl)-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-cyclohexylmethyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-benzyl-2-methyl-1H-imidazo[4,5-c]quinolin-4-amide;

sarcosine 1-n-hexyl-1H-imidazo[4,5-c]quinolin-4-amide; or

sarcosine 1-n-hexyl-2-methyl-1H-imidazo[4,5-c]quinolin-4-amide;

or a pharmaceutically acceptable salt thereof.
