Abstract

Hydrogels comprising a macromolecular matrix and water may be used to augment soft tissue of a human being, promote or support cell or tissue viability or proliferation, create space in tissue, and for other purposes. A macromolecular matrix may comprise a hyaluronic acid component crosslinked to a collagen component.

Claims (49)

1 . A soft tissue augmentation product comprising:

a forming component comprising a hydrogel having a form suitable for augmenting human soft tissue by injecting or implanting the forming component into the human tissue; and

a label comprising instructions to inject or implant the forming component into the human tissue;

wherein the hydrogel comprises water, and a crosslinked macromolecular matrix comprising:

a hyaluronic acid component; and

a collagen component;

wherein the hyaluronic acid component is crosslinked to the collagen component by an ester bond or an amide bond.

2 . The product of claim 1 , wherein crosslinked macromolecular matrix has a weight ratio of the hyaluronic acid component to the collagen component of about 1 to about 7.

3 . The product of claim 1 , wherein crosslinked macromolecular matrix has a weight ratio of the hyaluronic acid component to the collagen component of about 3.

4 . The product of claim 1 , wherein crosslinked macromolecular matrix has a weight ratio of the hyaluronic acid component to the collagen component of about 5.

5 . The product of claim 1 , wherein crosslinked macromolecular matrix has a weight ratio of the hyaluronic acid component to the collagen component of about 7.

6 . The product of claim 1 , wherein the hyaluronic acid is about 0.2% to about 2.5% of the weight of the hydrogel and the collagen component is about 0.05% to about 1.2% of the weight of the hydrogel.

7 . The product of claim 1 , wherein the hydrogel further comprises human adipose tissue.

8 . The product of claim 1 , wherein the hydrogel has a collagen concentration of about 3 mg/mL to about 12 mg/mL.

9 . The product of claim 1 wherein the hydrogel has a hyaluronic acid concentration of about 3 mg/mL to about 24 mg/mL.

10 . The product of claim 1 , wherein the hydrogel has a collagen concentration of at least about 3 mg/mL and a hyaluronic acid concentration of about 3 mg/mL to about 24 mg/mL.

11 . The product of claim 10 having a hyaluronic acid concentration of about 3 mg/mL.

12 . The product of claim 10 having a hyaluronic acid concentration of about 12 mg/mL.

13 . The product of claim 10 having a hyaluronic acid concentration of about 16 mg/mL.

14 . The product of claim 10 having a hyaluronic acid concentration of about 20 mg/mL.

15 . The product of claim 10 having a hyaluronic acid concentration of about 24 mg/mL.

16 . The product of claim 10 having a collagen concentration of at least about 6 mg/mL.

17 . The product of claim 16 having a hyaluronic acid concentration of about 12 mg/mL.

18 . The product of claim 16 having a hyaluronic acid concentration of about 16 mg/mL.

19 . The product of claim 16 having a hyaluronic acid concentration of about 20 mg/mL.

20 . The product of claim 16 having a hyaluronic acid concentration of about 24 mg/mL.

21 . The product of claim 10 having a collagen concentration of at least about 12 mg/mL, and a hyaluronic acid concentration of between about 12 mg/mL and about 24 mg/mL.

22 . A method of augmenting soft tissue of a human being comprising:

providing a hydrogel composition comprising water, and a crosslinked macromolecular matrix comprising:

a hyaluronic acid component, and

a collagen component;

wherein the hyaluronic acid component is crosslinked to the collagen component by a crosslinking component comprising a plurality of crosslink units, wherein at least a portion of the crosslink units comprises an ester bond or an amide bond; and

mixing the hydrogel composition with adipose tissue; and

introducing the hydrogel-adipose tissue mixture into soft tissue of the human being;

wherein the adipose tissue comprises cells, the cells including adipocytes, multiple types of regenerative cells, stromal vascular fraction cells, or a combination thereof.

The method of claim 22 , wherein crosslinked macromolecular matrix has a weight ratio of the hyaluronic acid component to the collagen component of about 1 to about 7, wherein the hyaluronic acid component is about 0.2% to about 2.5% of the weight of the hydrogel composition, and the collagen component is about 0.05% to about 1.2% of the weight of the hydrogel composition.

The method of claim 22 , wherein the hydrogel composition promotes or supports proliferation or survival of a cellular component in vivo to a greater extent than a hydrogel composition that is substantially identical except that the hyaluronic acid component and the collagen component are not crosslinked.

The method of claim 22 , wherein the hydrogel composition supports proliferation of stromal vascular fraction cells to a greater extent than a hydrogel composition comprising water and hyaluronic acid without a collagen, wherein the hyaluronic acid is present at a concentration of about 16 mg/mL.

The method of claim 22 , wherein the hydrogel composition allows diffusion of adipose tissue-specific growth factors to a greater extent than a hydrogel composition comprising water and hyaluronic acid without a collagen, wherein the hyaluronic acid is present at a concentration of about 16 mg/mL.

27 . The method of claim 22 , wherein the hydrogel composition allows diffusion of pro-angiogenic growth factors to a greater extent than a hydrogel composition comprising water and hyaluronic acid without a collagen, wherein the hyaluronic acid is present at a concentration of about 16 mg/mL.

28 . The method of claim 22 , wherein the hydrogel composition promotes adipogenesis to a greater extent than a hydrogel composition comprising water and hyaluronic acid without a collagen, wherein the hyaluronic acid is present at a concentration of about 16 mg/mL.

29 . The method of claim 22 , wherein the hydrogel composition promotes collagenesis to a greater extent than a hydrogel composition comprising water and hyaluronic acid without a collagen, wherein the hyaluronic acid is present at a concentration of about 16 mg/mL.

30 . A method of preparing a space in human or animal tissue comprising injecting a hydrogel composition into the tissue, wherein the hydrogel composition comprises water and a crosslinked macromolecular matrix comprising:

a hyaluronic acid component; and

a collagen component;

wherein the hyaluronic acid component is crosslinked to the collagen component by a crosslinking component; and

wherein the crosslinking component comprises a plurality of crosslink units, wherein at least a portion of the crosslink units comprise an ester bond or an amide bond.

31 . The method of claim 30 , wherein the hydrogel composition degrades in the tissue over a period of about 1 week to about 3 months to thereby create the space in the tissue.

32 . The method of claim 30 , further comprising injecting a human or animal fat composition into the space in the tissue.
