Regeneration of spinal discs
Compositions and processes for the effective and efficient regeneration of spinal discs are provided. These compositions contain stem cells, donor cells, and platelet plasma compositions. By using these compositions, there is an increased likelihood of acceptance and proper cell differentiation.
1. A process for regenerating a disc in a person in need of disc regeneration, said process comprising:
(a) harvesting bone marrow from the person;
(b) generating bone marrow aspirate and a platelet plasma composition from the bone marrow, wherein the bone marrow aspirate comprises mesenchymal stem cells, plasma, platelets and red blood cells;
(c) harvesting disc chondrocytes and an extracellular matrix from non-degenerative disc tissue of the person;
(d) combining the bone marrow aspirate and the disc chondrocytes to form a composition under conditions that permit the mesenchymal stem cells to begin differentiation through socialization due to cell to cell signaling; and
(e) administering the composition to a disc of the person in need of disc regeneration.
2. The process of claim 1 further comprising administering a scaffold to the person.
3. The process according to claim 2 , wherein the scaffold comprises at least one substance selected from the group consisting of platelet poor plasma, hyaluronic acid, fibrin glue and a flowable wound matrix.
4. The process according to claim 2 , wherein the scaffold comprises atelocollagen, hyaluronic acid and PEG-PLA.
5. The process of according to claim 1 further comprising mixing the chondrocytes, the extra cellular matrix, the bone marrow aspirate and the platelet plasma composition with stem cells derived from adipose tissue.
6. The process according to claim 5 , wherein the adipose tissue is subcutaneous fat.
7. The process according to claim 6 , wherein the mesenchymal stem cells from bone marrow aspirate and the stem cells from adipose tissue each contribute 40-60% of the total stem cells by volume.
8. The process according to claim 1 , wherein said step (c) harvesting comprises morselizing donor disc material.
9. The process according to claim 1 , wherein said step (c) harvesting comprises extruding the donor disc material.
10. The process of claim 1 further comprising separating blood plasma into platelet rich plasma and platelet poor plasma, and exposing the platelet rich plasma to conditions that lyse membranes of cells in the platelet rich plasma to form a lysate, wherein the platelet plasma composition comprises the lysate.
11. The process according to claim 1 , wherein the composition further comprises LIM mineralization protein (LMP)-1.
12. The process according to claim 1 , wherein said combining further comprises adding calcium chloride.
13. The process according to claim 12 , wherein said combining further comprises adding thrombin.
14. The process according to claim 1 , wherein said combining further comprises adding insulin-like growth factor-1.
15. The process according to 1 , wherein said combining further comprises adding bone morphogenic protein.
16. The process according to claim 1 , wherein said combining further comprises adding Sox-9.
17. The process according to claim 1 , wherein said combining further comprises adding genipin.
18. The process according to claim 1 , further comprising photo-activating the composition.
19. The process according to claim 18 , wherein said photo-activating is under conditions that increase the presence of Interleukin-1 Receptor Antagonist.
