Method for controlling the amount of gene product, and agent for controlling the amount of gene product
The invention relates to a method of intracellularly controlling amounts of gene products, which can increase an amount of gene product intracellularly, comprising a step of introducing into the cell a substance having a sequence complementary to the base sequence of mRNA corresponding to the gene product, its precursor or another substance which can have equivalent action in the cell.
1. A method for identifying a protein that can be overexpressed by introducing an antisense polynucleotide into a cell that expresses the protein, said method comprising:
(a) screening RNA molecules from one or more cells to identify an antisense transcript that contains a nucleotide sequence complementary to a 3′-untranslated region (UTR) of an mRNA that encodes an early-response protein, said mRNA being present in the one or more cells;
(b) introducing, into a cell expressing said mRNA, an antisense polvnucleotide containing a nucleotide sequence complementary to the 3′-untranslated region (UTR) of said mRNA, and measuring whether the introduction of said antisense polynucleotide increases expression of the early-response protein in the cell; and
(c) determining that said mRNA encodes a protein which can be overexpressed by introducing an antisense polynucleotide containing a nucleotide sequence complementary to the 3′-untranslated region (UTR) of said mRNA into a cell expressing said early-response protein, when an increase in expression of the early-response protein in the cell is measured in step (b).
2. The method of claim 1 , wherein the identification is performed by reverse transcribing RNA from one or more cells using a primer containing a sense sequence from the 5′-or 3′-end of an mRNA, and detecting the presence of a reverse-transcription product.
