Abstract

The invention relates to compositions and methods for treating kidney and muscle dysfunction that involves use of therapeutic amounts of N-acetyl mannosamine.

Claims (13)

1. A method of treating a kidney disease or condition in a mammal in need thereof comprising administering an effective amount of a sialic acid precursor, N-acetyl mannosamine or a derivative thereof, to the mammal to thereby treat a kidney disease or condition in the mammal, wherein the derivative consists of Formula I;

wherein: R 1 , R 3 , R 4 , or R 5 is hydrogen, lower alkanoyl, carboxylate or lower alkyl; and R 2 is lower alkyl, lower alkanoylalkyl, or lower alkyl alkanoyloxy, the kidney disease or condition is selected from podocytopathy, minimal change nephrosis, focal and segmental glomerulosclerosis, membranous glomerulonephritis, glomerular basement membrane disease, nephrotic syndrome, IgA nephropathy, Alport disease, thin membrane disease, reflux nephropathy, or other kidney disease or condition with hyposialylation, and the administration is rectal, dermal, transdermal, intrathoracic, intrapulmonary, intranasal, subcutaneous, intravenous, or intramuscular.

2. The method of claim 1 , comprising administering a therapeutic amount of N-acetyl mannosamine to the mammal.

A method of treating a kidney disease or condition in a mammal in need thereof comprising sustained release administering an effective amount of a sialic acid precursor, N-acetyl mannosamine or a derivative thereof, to the mammal to thereby treat a kidney disease or condition in the mammal, wherein the derivative consists of Formula I;

wherein: R 1 , R 3 , R 4 , or R 5 is hydrogen, lower alkanoyl, carboxylate or lower alkyl; and R 2 is lower alkyl, lower alkanoylalkyl, or lower alkyl alkanoyloxy, the kidney disease or condition is selected from podocytopathy, minimal change nephrosis, focal and segmental glomerulosclerosis, membranous glomerulonephritis, glomerular basement membrane disease, nephrotic syndrome or other kidney disease or condition with hyposialylation.

4. The method of claim 3 , comprising administering a therapeutic amount of N-acetyl mannosamine to the mammal.

A method of treating a kidney disease or condition in a mammal in need thereof comprising topically administering an effective amount of a sialic acid precursor, N-acetyl mannosamine or a derivative thereof, to the mammal to thereby treat a kidney disease or condition in the mammal, wherein the derivative consists of Formula I;

wherein: R 1 , R 3 , R 4 , or R 5 is hydrogen, lower alkanoyl, carboxylate or lower alkyl; and R 2 is lower alkyl, lower alkanoylalkyl, or lower alkyl alkanoyloxy, the kidney disease or condition is selected from podocytopathy, minimal change nephrosis, focal and segmental glomerulosclerosis, membranous glomerulonephritis, glomerular basement membrane disease, nephrotic syndrome or other kidney disease or condition with hyposialylation.

6. The method of claim 5 , comprising administering a therapeutic amount of N-acetyl mannosamine to the mammal.

7. A method of treating a condition or disease in a mammal in need thereof comprising administering orally an effective amount of a sialic acid precursor, N-acetyl mannosamine or a derivative thereof, to the mammal, wherein the derivative consists of Formula I

wherein R 1 , R 3 , R 4 , or R 5 is hydrogen, lower alkanoyl, carboxylate or lower alkyl; R 2 is lower alkyl, lower alkanoylalkyl, lower alkyl alkanoyloxy; and the condition or disease is selected from IgA nephropathy, Alport disease, thin membrane disease, or reflux nephropathy.

8. The method of claim 3 , wherein the sustained release administering comprises administering via an indwelling device.

9. The method of claim 5 , wherein topically administering comprises administering via a cream, gel, lotion, impregnated pad, ointment, aerosol formulation, soap, polymer covering, or drops.
