Abstract

The present disclosure provides antibodies that specifically bind to botulinum neurotoxins (e.g., BoNT/A, BoNT/B, BoNT/C, BoNT/D, BoNT/E, BoNT/F, BoNT/G, etc.) and the epitopes bound by those antibodies. The antibodies and derivatives thereof that specifically bind to the neutralizing epitopes provided herein can be used to neutralize botulinum neurotoxin and are therefore also useful in the treatment of botulism.

Claims (56)

1. An isolated antibody, or antigen-binding fragment thereof, that competes for binding with, or specifically binds an epitope of a Botulinum neurotoxin that is specifically bound by, an antibody comprising:

a) a V H CDR1 comprising the amino acid sequence of residues 31-35 of SEQ ID NO:68;

b) a V H CDR2 comprising the amino acid sequence of residues 50-66 of SEQ ID NO:68;

c) a V H CDR3 comprising the amino acid sequence of residues 99-114 of SEQ ID NO:68;

d) a V L CDR1 comprising the amino acid sequence of residues 24-34 of SEQ ID NO:205;

e) a V L CDR2 comprising the amino acid sequence of residues 50-56 of SEQ ID NO:205; and

f) a V L CDR3 comprising the amino acid sequence of residues 89-97 of SEQ ID NO:205.

2. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody, or antigen-binding fragment thereof, specifically binds an epitope of a Botulinum neurotoxin that is specifically bound by an antibody comprising:

a) a V H CDR1 comprising the amino acid sequence of residues 31-35 of SEQ ID NO:68;

b) a V H CDR2 comprising the amino acid sequence of residues 50-66 of SEQ ID NO:68;

c) a V H CDR3 comprising the amino acid sequence of residues 99-114 of SEQ ID NO:68;

d) a V L CDR1 comprising the amino acid sequence of residues 24-34 of SEQ ID NO:205;

e) a V L CDR2 comprising the amino acid sequence of residues 50-56 of SEQ ID NO:205; and

f) a V L CDR3 comprising the amino acid sequence of residues 89-97 of SEQ ID NO:205.

3. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody, or antigen-binding fragment thereof, is a humanized antibody.

4. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody is a single chain Fv (scFv), IgG, Fab, (Fab) 2 , or (scFv′) 2 .

5. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody, or antigen-binding fragment thereof, is a human monoclonal antibody.

6. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody, or antigen-binding fragment thereof, comprises:

a) a V H CDR1 comprising the amino acid sequence of residues 31-35 of SEQ ID NO:68;

b) a V H CDR2 comprising the amino acid sequence of residues 50-66 of SEQ ID NO:68;

c) a V H CDR3 comprising the amino acid sequence of residues 99-114 of SEQ ID NO:68;

d) a V L CDR1 comprising the amino acid sequence of residues 24-34 of SEQ ID NO:205;

e) a V L CDR2 comprising the amino acid sequence of residues 50-56 of SEQ ID NO:205; and

f) a V L CDR3 comprising the amino acid sequence of residues 89-97 of SEQ ID NO:205.

7. The isolated antibody, or antigen-binding fragment thereof, of claim 1 , wherein said antibody competes for binding to a Botulinum neurotoxin with an antibody comprising:

a) a V H CDR1 comprising the amino acid sequence of residues 31-35 of SEQ ID NO:68;

b) a V H CDR2 comprising the amino acid sequence of residues 50-66 of SEQ ID NO:68;

c) a V H CDR3 comprising the amino acid sequence of residues 99-114 of SEQ ID NO:68;

d) a V L CDR1 comprising the amino acid sequence of residues 24-34 of SEQ ID NO:205;

e) a V L CDR2 comprising the amino acid sequence of residues 50-56 of SEQ ID NO:205; and

f) a V L CDR3 comprising the amino acid sequence of residues 89-97 of SEQ ID NO:205.

8. A composition comprising:

a pharmaceutically acceptable carrier; and

the antibody, or antigen-binding fragment thereof, according to claim 1 .

9. A kit comprising a composition of claim 8 .

10. A method for detecting Botulinum neurotoxin in a sample, comprising:

contacting an antibody, or antigen-binding fragment thereof, of claim 1 with the sample,

detecting binding of said antibody to Botulinum neurotoxin in said sample.

11. The method of claim 10 , wherein said antibody is labeled.

12. The method of claim 11 , wherein said label is a fluorescent label, a chemiluminescent label, a radiolabel, an enzyme, or a chromogenic label.

13. The method of claim 10 , wherein said sample is blood.

14. The method of claim 10 , wherein said antibody is immobilized on a substrate.

15. The method of claim 10 , wherein said antibody is conjugated to a nucleic acid and wherein said detecting comprises a polymerase chain reaction.

16. A method of reducing the likelihood that an individual at risk of exposure to Botulinum neurotoxin will experience symptoms of Botulinum neurotoxin poisoning following exposure, the method comprising administering to said individual an effective amount of an antibody of claim 1 ;

wherein said administering provides for reducing the likelihood that the individual will experience symptoms of Botulinum neurotoxin poisoning.

17. The isolated antibody or antigen-binding fragment of claim 1 , wherein said antibody comprises:

a heavy chain comprising the amino acid sequence of SEQ ID NO:68.

18. The isolated antibody or antigen-binding fragment of claim 1 , wherein said antibody comprises:

a light chain comprising the amino acid sequence of SEQ ID NO:205.

19. The isolated antibody or antigen-binding fragment of claim 1 , wherein said antibody comprises:

a) a heavy chain comprising the amino acid sequence of SEQ ID NO:68; and

b) a light chain comprising the amino acid sequence of SEQ ID NO:205.

20. The isolated antibody or antigen-binding fragment of claim 19 , wherein said antibody is a human monoclonal antibody.

21. The isolated antibody or antigen-binding fragment of claim 19 , wherein said antibody is a single chain Fv (scFv), Fab, (Fab′) 2 or (scFv′) 2 .

22. The isolated antibody or antigen-binding fragment of claim 1 , wherein said antibody is an IgG.

23. The isolated antibody or antigen-binding fragment of claim 19 , wherein said antibody is an IgG.
