Abstract

The present invention relates to compositions comprising: a polypeptide having cellulolytic enhancing activity and a quinone compound. The present invention also relates to methods of using the compositions.

Claims (52)

1. A method for degrading or converting a cellulosic material, comprising: saccharifying the cellulosic material with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity and a quinone compound, wherein the combination of the GH61 polypeptide having cellulolytic enhancing activity and the quinone compound enhances hydrolysis of the cellulosic material by the enzyme composition compared to the GH61 polypeptide alone or the quinone compound alone according to the following formula with a ratio greater than 1:

2. The method of claim 1 , further comprising recovering the degraded or converted cellulosic material.

3. The method of claim 1 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin.

4. The method of claim 1 , wherein the quinone compound is a compound of formula (I) or (II):

wherein R 1 , R 2 , R 3 , and R 4 are independently hydrogen, halogen, —OH, —OR 8 , —NO 2 , —N(R 9 )(R 10 ), —CN, —C(O)R 5 , —C(O)OR 6 , —C(O)NHR 7 , —OC(O)R 11 , —NHC(O)R 12 , —OC(O)OR 13 , —NHC(O)OR 14 , —OC(O)NHR 15 , —NHC(O)NHR 16 , —SO 2 R 17 , —SO 2 N(R 18 )(R 19 ), —SR 20 , —NC(R 21 )(R 22 ), or an optionally substituted moiety selected from alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkyl-alkyl, heterocycloalkyl, heterocycloalkyl-alkyl, aryl, aralkyl, heteroaryl, and heteroaralkyl;

R 5 , R 6 , R 7 , R 8 , R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , R 18 , R 19 , R 20 , R 21 , and R 22 are independently hydrogen, or an optionally substituted moiety selected from alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkyl-alkyl, heterocycloalkyl, heterocycloalkyl-alkyl, aryl, aralkyl, heteroaryl, and heteroaralkyl; and

R 17 is an optionally substituted moiety selected from alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkyl-alkyl, heterocycloalkyl, heterocycloalkyl-alkyl, aryl, aralkyl, heteroaryl, and heteroaralkyl; and

wherein each pair of R 1 and R 2 , R 2 and R 3 , and R 3 and R 4 may combine to form an optionally substituted fused ring;

or a salt or solvate thereof.

The method of claim 4 , wherein the quinone compound is selected from the group consisting of: (I-1): 1,4-benzoquinone; (I-2): 1,4-naphthoquinone; (I-3): 2-hydroxy-1,4-naphthoquinone; (I-4): 2,3-dimethoxy-5-methyl-1,4-benzoquinone or coenzyme Q 0 ; (I-5): 2,3,5,6-tetramethyl-1,4-benzoquinone or duroquinone; (I-6): 1,4-dihydroxyanthraquinone; (II-1): 3-hydroxy-1-methyl-5,6-indolinedione or adrenochrome; (II-2): 4-tert-butyl-5-methoxy-1,2-benzoquinone; and (II-3): pyrroloquinoline quinone; or a salt or solvate thereof.

6. The method of claim 1 , wherein the cellulosic material is pretreated.

7. The method of claim 3 , wherein the cellulase is one or more enzymes selected from the group consisting of an endoglucanase, a cellobiohydrolase, and a beta-glucosidase.

8. The method of claim 3 , wherein the hemicellulase is one or more enzymes selected from the group consisting of a xylanase, an acetyxylan esterase, a feruloyl esterase, an arabinofuranosidase, a xylosidase, and a glucuronidase.

9. The method of claim 1 , wherein the degraded or converted cellulosic material is a sugar.

10. The method of claim 9 , wherein the sugar is selected from the group consisting of glucose, xylose, mannose, galactose, and arabinose.

11. The method of claim 1 , wherein an effective amount of the quinone compound to cellulosic material as a molar ratio to glucosyl units of cellulose is about 10 −6 to about 10.
