Abstract

Bioactive porous bone graft implants in various forms suitable for bone tissue regeneration and/or repair, as well as methods of use, are provided. The implants are formed of bioactive glass and have an engineered porosity. The implants may take the form of a putty, foam, fibrous cluster, fibrous matrix, granular matrix, or combinations thereof and allow for enhanced clinical results as well as ease of handling.

Claims (33)

1. A porous, composite bone graft implant comprising:

a first component comprising a bioactive glass material in the form of a plurality of glass fibers; and

a second component comprising a bioactive glass material in the form of a coating over the plurality of fibers;

each of the components having a different resorption capacity than the other component;

wherein the composite implant comprises a pore size distribution including pores characterized by pore diameters ranging from about 100 nanometers to about 1 millimeter.

2. The implant of claim 1 , wherein at least one of the first and second components is partially or fully sintered.

3. The implant of claim 1 , wherein at least one of the first and second components is porous.

4. The implant of claim 1 , wherein the second component coating is porous.

5. The implant of claim 1 , wherein the plurality of fibers further comprises bioactive glass granules.

6. The implant of claim 5 , wherein the coating extends over all of the plurality of fibers and granules.

The implant of claim 1 , wherein the bioactive glass material is selected from the group consisting of sol gel derived bioactive glass, melt derived bioactive glass, silica based bioactive glass, silica free bioactive glass, phosphate based bioactive glass, crystallized bioactive glass, and bioactive glass containing trace elements.

8. The implant of claim 7 , wherein the bioactive glass material comprises a silica free bioactive glass selected from the group consisting of borate based bioactive glass and phosphate based bioactive glass.

9. The implant of claim 7 , wherein the bioactive glass material comprises a partially crystallized or fully crystallized bioactive glass.

10. The implant of claim 1 , further comprising a carrier material.

The implant of claim 10 , wherein the carrier material comprises human-derived collagen, animal-derived collagen, phospholipids, carboxylmethylcellulose (CMC), glycerin, polyethylene glycol (PEG), polylactic acid (PLA), polylactic-co-glycolic acid (PLGA), or other copolymers of the same family.

12. The implant of claim 10 , wherein the carrier material comprises hyaluronic acid, sodium alginate, saline, or bone marrow aspirate.

13. A porous, composite bone graft implant comprising:

a first component comprising a bioactive glass material in the form of a plurality of granules; and

a second component comprising a bioactive material in the form of a coating over the plurality of granules;

each of the components having a different resorption capacity than the other component;

wherein the composite implant comprises a pore size distribution including pores characterized by pore diameters ranging from about 100 nanometers to about 1 millimeter.

14. The implant of claim 13 , wherein the first component further comprises bioactive glass fibers.

15. The implant of claim 13 , wherein the coating comprises a bioactive glass or glass-ceramic.

16. The implant of claim 13 , wherein at least one of the first and second components is partially or fully sintered.

17. The implant of claim 13 , wherein at least one of the first and second components is porous.

18. The implant of claim 13 , wherein the coating is porous.

19. The implant of claim 13 , wherein each of the granules comprises a further coating comprising bioactive glass.

The implant of claim 13 , wherein the bioactive glass material comprises sol gel derived bioactive glass, melt derived bioactive glass, silica based bioactive glass, silica free bioactive glass, or phosphate based bioactive glass, partially crystallized bioactive glass, fully crystallized bioactive glass, or bioactive glass containing trace elements.

21. The implant of claim 20 , wherein the bioactive glass material comprises a silica free bioactive glass selected from the group consisting of borate based bioactive glass and phosphate based bioactive glass.

22. The implant of claim 20 , wherein the bioactive glass material comprises a partially crystallized or fully crystallized bioactive glass.

23. The implant of claim 13 , further comprising a carrier material.

The implant of claim 23 , wherein the carrier material comprises human-derived collagen, animal-derived collagen, phospholipids, carboxylmethylcellulose (CMC), glycerin, polyethylene glycol (PEG), polylactic acid (PLA), polylactic-co-glycolic acid (PLGA), or other copolymers of the same family.

25. The implant of claim 23 , wherein the carrier material comprises hyaluronic acid, sodium alginate, saline, or bone marrow aspirate.
