Methods and compositions using miR-3151 in the diagnosis and treatment of cancer
Compositions comprising therapeutic oligonucleotide compounds that target the expression of genes associated with tumorigenesis or cell transformation are provided. Methods are described to manipulate and treat the oncogenic deregulation of the TP53-associated apoptosis pathway. Markers for acute myeloid leukemia (AML) and uses thereof are described.
1. A method of treating a subject having or suspected of having a cancer having an increased expression of miR-3151, the method comprising:
administering to the subject an effective amount of a miR-3151 inhibitor sufficient to modulate expression of miR-3151, wherein the levels of miR-3151 are decreased after administration, thereby treating the subject;
wherein the cancer is cytogenetically normal acute myeloid leukemia (CN-AML).
2. A method of treating a patient having acute myelogenous leukemia (AML) or acute lymphoblastic leukemia (ALL) comprising:
measuring the expression of at least one biomarker in a biological sample from a patient, wherein the at least one biomarker is selected from the group consisting of: gene products of BAALC and miR-3151;
comparing the expression of the at least one biomarker to a corresponding control;
determining whether the expression of either BAALC or miR-3151 is high;
administering antagomiR-3151 if the expression of either BAALC or miR-3151 is high, in an amount sufficient to modulate expression of one or more of BAALC and miR-3151; wherein the levels of one or more of BAALC and miR-3151 are decreased after administration, thereby treating the subject.
3. The method of claim 2 further comprising: monitoring the expression of the at least one biomarker subsequent to the step of administering the antagomiR-3151.
4. A method of inhibiting miR-3151 expression in a subject undergoing treatment for acute myeloid leukemia (AML) comprising:
administering to a subject undergoing treatment for ALL, in which at least miR-3151 gene product is upregulated in cancer cells of the subject, an effective amount of at least one compound for inhibiting expression of the at least one miR-3151 gene product in an amount sufficient to modulate expression of miR-3151; wherein the levels of miR-3151 are decreased after administration, thereby inhibiting miR-3151 expression in the subject.
5. A method of inhibiting a leukemia in a subject who has acute myeloid leukemia (AML), comprising:
determining the amount of at least one miR-3151 gene product in cancer cells from a subject;
altering the amount of miR-3151 gene product expressed in the cancer cells by administering to the subject an effective amount of at least one compound for inhibiting expression of the at least one miR-3151 gene product, if the amount of the miR-3151 gene product expressed in the cancer cells is greater than the amount of the miR-3151 gene product expressed in control cells; and
inhibiting AML in the subject.
6. The method of claim 5 wherein the compound comprises antagomiR-3151.
