Natural killer cells with enhanced immune response
View Patent ↗The invention relates to a specialized subpopulation of natural killer cells that have enhanced effector functions and the potential to kill malignant tumor cells or infected cells when the natural killer cells are exposed to an antibody bound to the tumor cells or the infected cells.
1. A method of treating a subject having cancer, comprising administering to the subject a composition comprising a cell population comprising an effective amount of natural killer cells that do not express substantial FcRγ (g − NK cells), and administering one or more therapeutic antibody that binds to the cancer cells to the subject separately or with the composition comprising g − NK cells, wherein the one or more therapeutic antibody binds to CD16 on the surface of the g − NK cells, to thereby treat the cancer in the subject.
2. The method of claim 1 , wherein the g − NK cells in the composition comprising g − NK cells are isolated from the subject.
3. The method of claim 2 , wherein the g − NK cells in the composition comprising g − NK cells isolated from the subject are cultured in a cell culture medium prior to administering the composition.
4. The method of claim 2 , wherein IL-12, IL-15, IL-18, IL-2, and/or CCL5 is administered to the subject prior to isolating the g − NK cells.
5. The method of claim 1 , further comprising identifying whether the subject has g − NK cells by identifying cells that do not express substantial FcRγ, but do express at least one marker for natural killer cells, prior to administering the composition.
6. The method of claim 5 , wherein identifying whether the subject has g − NK cells comprises obtaining lymphocytes from the subject.
7. The method of claim 1 , wherein the g − NK cells express KIR2DL2/3, or NKG2C, or a combination thereof.
8. The method of claim 1 , wherein the cell population comprises at least 70% g − NK cells.
9. The method of claim 1 , wherein the g − NK cells express low levels of NKp30 and NKp46, compared to conventional NK cells.
10. The method of claim 1 , wherein the g − NK cells in the composition comprising g − NK cells are isolated from a donor.
11. The method of claim 10 , wherein the g − NK cells in the composition comprising g − NK cells isolated from the donor are cultured in a cell culture medium prior to administering the composition.
12. The method of claim 10 , wherein IL-12, IL-15, IL-18, IL-2, and/or CCL5 is administered to the donor prior to isolating the g − NK cells.
