Abstract

The present application relates to anti-LOX and anti-LOXL2 antibodies and their use in purification, diagnostic and therapeutic methods. Antibodies include monoclonal antibodies, humanized antibodies and functional fragments thereof. Anti-LOX and anti-LOXL2 antibodies can be used to identify and treat conditions such as a fibrotic condition, angiogenesis, or to prevent a transition from an epithelial cell state to a mesenchymal cell state.

66 . An isolated antibody or antigen binding fragment thereof, comprising a heavy chain variable region having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 25, 26, 27 or 28.

67 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the heavy chain variable region comprises at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 27.

68 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the heavy chain variable region comprises at least 95% identity to the amino acid sequence set forth as SEQ ID NO: 25, 26, 27 or 28.

69 . The isolated antibody or antigen binding fragment thereof of claim 68 , wherein the heavy chain variable region comprises at least 99% identity to the amino acid sequence set forth as SEQ ID NO: 25, 26, 27 or 28.

70 . The isolated antibody or antigen binding fragment of claim 66 , further comprising a light chain variable region having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 30, 31, or 32.

71 . The isolated antibody or antigen binding fragment of claim 70 , wherein the light chain variable region has at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 32.

72 . The isolated antibody or antigen binding fragment of claim 70 , wherein the light chain variable region has at least 95% identity to the amino acid sequence set forth as 30, 31, or 32.

73 . The isolated antibody or antigen binding fragment of claim 72 , wherein the light chain variable region has at least 99% identity to the amino acid sequence set forth as 30, 31, or 32.

74 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the antibody or fragment is labeled with a detectable label, a therapeutic agent, or both.

75 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the antibody or fragment is labeled with a therapeutic agent.

76 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the antibody or fragment is a Fv, a scFv, a Fab, a F(ab′) 2 , or monoclonal.

77 . The isolated antibody or antigen binding fragment thereof of claim 66 , wherein the antibody or fragment is humanized.

78 . An isolated antibody or antigen binding fragment thereof, comprising:

a heavy chain variable region having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 25, 25, 26, 27 or 28; and

a light chain variable region having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 30, 31, or 32,

wherein the isolated antibody or antigen binding fragment thereof specifically binds to a LOXL2 protein.

79 . The isolated antibody or antigen binding fragment of claim 78 , wherein the heavy chain variable region has at least 95% identity to the amino acid sequence set forth as SEQ ID NO: 25, 25, 26, 27 or 28 and the light chain variable region has at least 95% identity to the amino acid sequence set forth as SEQ ID NO: 30, 31, or 32.

80 . The isolated antibody or antigen binding fragment of claim 79 , wherein the heavy chain variable region has at least 99% identity to the amino acid sequence set forth as SEQ ID NO: 25, 25, 26, 27 or 28 and the light chain variable region has at least 99% identity to the amino acid sequence set forth as SEQ ID NO: 30, 31, or 32.

81 . The isolated antibody or antigen binding fragment thereof of claim 78 , wherein the antibody or fragment is labeled with a detectable label, a therapeutic agent, or both.

82 . The isolated antibody or antigen binding fragment thereof of claim 78 , wherein the antibody or fragment is labeled with a therapeutic agent.

83 . The isolated antibody or antigen binding fragment thereof of claim 78 , wherein the antibody or fragment is a Fv, a scFv, a Fab, a F(ab′) 2 , or monoclonal.

84 . The isolated antibody or antigen binding fragment thereof of claim 78 , wherein the antibody or fragment is humanized.

85 . An isolated antibody or antigen binding fragment thereof, comprising:

a variable heavy chain having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 27; and

a variable light chain having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 32,

wherein the isolated antibody or antigen binding fragment thereof specifically binds to a LOXL2 protein.

86 . The isolated antibody or antigen binding fragment thereof of claim 79 , wherein the heavy chain variable region comprises at least 95% identity to the amino acid sequence set forth as SEQ ID NO:27 and the light chain variable region comprises at least 95% identity to the amino acid sequence set forth as SEQ ID NO:31.

87 . The isolated antibody or antigen binding fragment thereof of claim 86 , wherein the heavy chain variable region comprises at least 99% identity to the amino acid sequence set forth as SEQ ID NO:27 and the light chain variable region comprises at least 99% identity to the amino acid sequence set forth as SEQ ID NO:31.

88 . The isolated antibody or antigen binding fragment thereof of claim 85 , wherein the antibody or fragment is labeled with a detectable label, a therapeutic agent, or both.

89 . The isolated antibody or antigen binding fragment thereof of claim 85 , wherein the antibody or fragment is labeled with a therapeutic agent.

90 . The isolated antibody or antigen binding fragment thereof of claim 85 , wherein the antibody or fragment is a Fv, a scFv, a Fab, a F(ab′) 2 , or monoclonal.

91 . The isolated antibody or antigen binding fragment thereof of claim 85 , wherein the antibody or fragment is humanized.

92 . A pharmaceutical composition comprising an isolated antibody or antigen binding fragment thereof and a pharmaceutical acceptable carrier, wherein the isolated antibody or antigen binding fragment thereof comprising:

a variable heavy chain having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 25, 25, 26, 27 or 28; and a variable light chain having at least 90% identity to the amino acid sequence set forth as SEQ ID NO: 30, 31, or 32.

93 . The pharmaceutical composition of claim 92 , wherein composition may be administered intravenously, intraarterially, intramuscularly, subcutaneously, orally, or topically.
