Abstract

The present invention provides novel aspirin derivatives useful for preventing and/or treating cancer. The novel compounds of this invention may be particularly useful for the prevention and/or treatment of cancers affecting the gastrointestinal system, such as colorectal cancer.

Claims (50)

1. A compound having the following structure

or a pharmaceutically acceptable salt or prodrug thereof,

wherein:

E is —O—;

each of R A and R C is independently —H or —OH;

each of R D and R B is independently —H, —OH or —O—C 1-6 alkyl; and

R E is —O—C 1-6 acyl.

2. The compound of claim 1 , wherein R A is —OH.

3. The compound of claim 1 , wherein said compound has the structure

wherein:

each of R B and R D is independently —OH or —O—C 1-6 alkyl; and

R E is —O—C(O)-Me.

4. The compound of claim 3 , wherein each of R B and R D is —OH.

5. The compound of claim 3 , wherein each of R B and R D is independently —O—C 1-6 alkyl.

6. The compound of claim 3 , wherein each of R B and R D is independently —O—C 1-3 alkyl.

7. The compound of claim 3 , wherein each of R B and R D is —O-Me.

8. The compound of claim 1 , wherein said compound has the structure

wherein:

R A is —OH;

R C is —H or —OH; and

R E is —O—C 1-6 acyl.

9. The compound of claim 8 , wherein R C is —OH and R E is —O—C(O)Me.

10. A compound of Formula IV or Formula V

or a pharmaceutically acceptable salt or prodrug thereof.

11. A pharmaceutical composition, comprising:

a compound having the following structure

or pharmaceutically acceptable salt thereof; and

a pharmaceutically acceptable carrier,

wherein:

E is —O—;

each of R A and R C is independently —H or —OH;

each of R D and R B is independently —H, —OH or —O—C 1-6 alkyl; and

R E is —O—C 1-6 acyl.

12. The pharmaceutical composition of claim 11 , wherein said compound has the following structure

wherein:

each of R B and R D is independently —OH or —O—C 1-6 alkyl; and

R E is —O—C(O)-Me.

13. The pharmaceutical composition of claim 12 , wherein each of R B and R D is —OH.

14. The pharmaceutical composition of claim 12 , wherein each of R B and R D is —O-Me.

15. The pharmaceutical composition of claim 11 , wherein said compound has the following structure

wherein:

R A is —OH;

R C is —H or —OH; and

R E is —O—C 1-6 acyl.

16. The pharmaceutical composition of claim 15 , wherein R C is —OH and R E is —O—C(O)Me.

17. A pharmaceutical composition, comprising:

a compound of Formula IV or Formula V

or a pharmaceutically acceptable salt or prodrug thereof; and

a pharmaceutically acceptable carrier.

18. A compound having the following structure:
