Amylases, nucleic acids encoding them and methods for making and using them
In one aspect, the invention is directed to polypeptides having an amylase activity, polynucleotides encoding the polypeptides, and methods for making and using these polynucleotides and polypeptides. The polypeptides of the invention can be used as amylases to catalyze the hydrolysis of starch into sugars.
1. A variant polynucleotide comprising a nucleic acid sequence that is at least 95% identical to the full length nucleic acid sequence as set forth in SEQ ID NO:15, and the variant polynucleotide is operably linked to a heterologous promoter wherein the variant polynucleotide encodes a polypeptide having glucoamylase activity and the polypeptide is the amino acid sequence of SEQ ID NO: 16.
2. An expression cassette comprising the nucleic acid sequence of claim 1 .
3. A vector comprising the nucleic acid sequence of claim 1 .
4. A method of producing a recombinant polypeptide comprising:
(a) providing the nucleic acid operably linked to a promoter of claim 1 ; and
(b) expressing the nucleic acid of step (a) under conditions that allow expression of the polypeptide, thereby producing a recombinant polypeptide.
5. A method of generating a variant of the nucleic acid of claim 1 , comprising:
(a) providing a template nucleic acid as set forth in claim 1 ;
(b) modifying, deleting, or adding one or more nucleotides in the template sequence, to generate a variant of the template nucleic acid;
(c) expressing the variant of (b) to generate a variant glucoamylase.
